This article states about the experiment that used H.caput-medusae(Bull.:Fr.) Pers.liquid submerged fermentation extractum as crude materials and did some preliminary researches-such as isolation,purification, physico-chemical property,stability,biological activity and so on-abont oligosaccharide(HPY).And here came the following conclusions.1 By adopted the method of ethanol precipitation to process HPY,crude sugar yield was 15.5%.After acidation,repeated freeze thawing,millipore filter process respectively,and through column chromatography Q-Sepharose FF,then carried out the elution of polysaccharide at 3 ionic strengths.The result manifests that polysaccharide contained mostly neutral non-protein oligosaccharide.Under the optimal technology and craftsmanship,the application of negative and positive ion macroporous resin cascade connection adsorbed to the separation of non-protein oligosaccharide in narrow spectrum,losing rate of saccharide is 50.5%. Therefore,total yield of polysaccharides was 7.89%.The technology and craftsmanship H.caput-medusae (Bull.:Fr.) Pers.liquid submerged fermentation extractum oligosaccharide supplied sufficient raw materials for fungus oligosaccharide medication.2 By adopted the Phenol-Sulphuric Acid method,in H.caput-medusae(Bull.:Fr.) Pers liquid submerged fermentation extractum purified oligosaccharide(HPY-1),the estimated ratio of total sugar weight was 97.59%.The estimating method was stable,repeatable,thermally stable.The physical and chemical methods and Superdex 30p gel column chromatographic analysis leading to the result that HPY-1 was non-reducible single composition neutral oligosaccharide.Ultraviolet absorption spectrum analytic result manifests that HPY-1 was of no nucleonic acid and protein absorption.Infrared spectrum shows that HPY-1 contains the characteristic absorption peak and the glucosidal bond's configuration wasβ.And the inherent viscosity was[η]=45.The acetylation and GC analysis of HPY-1 showed monosaccharide composition was mainly glucose.3 During the research of anti-oxidation,the comparative analysis about HPY's和HPY-1's reducing power,superoxide anion radical removability and hydroxide free radical removability respectively.The result showed that the reducibility increases along with increasing density.When the density was 10mg/ml,the reducibility of HPY-1 reaches as much as 1.202.If in the same density,the reducibility of HPY-1 is obviously better than HPY.By adopting the pyrogallic acid method,the estimated superoxide anion clearance rate is HPY-1.3(33.6%)>HPY-1.2(25.9%)>HPY3(19.3%)>HPY2(15.6%)>HPY-1.1(11.5%)>HPY1 (7.9%).Similarly,in the same density,superoxide anion clearance ability of HPY-1 was obviously better than that of HPY.Concerning about the identical sample,superoxide anion clearance rate increases when the sample density increases.When the density was 0.4mg/ml,the clearance rate of HPY-1 reach 33.6%.The experiment,which adopted the method of removing hydroxide free radical,indicated the hydroxide free radical clearance rate was HPY-1.3(46.72%)>HPY3(31.52%)>HPY-1.2(29.48%)>HPY2(17.93%)>HPY-1.1(7.51%)>HPY1(3.58%).4 The experiment,which evaluated Hela and SMMC-7721 tumor cell's extraorgan IC50 through MTT, indicated 24-hour affection of HPY considerably inhibits the proliferation of chosen tumor cell.The highest inhibit rate reached more than 70%on average.In conclusion,HPY-1 will obviously inhibit the proliferation of Hela and SMMC-7721,and the did-affection relationship is satisfactory.
