In the paper,anther of six blueberry species is selected as the sample for in vitro culture study,and the factors affecting the induction of anther callus in dedifferentiation and the best suitable varieties and concentration with different ratios of culture medium for proliferation and differentiation in callus redifferentiation are intensively studied,and tissue culture seedlings of blueberry induced with colchicine and purification of the mutated blueberry plants are both researched.The results are as follows:1,Environmental conditions have important effects on induction of anther callus in differentiation.In light treatment,dark helps the induction of callus of blueberry anther,in which the induction rate reaches 53.33%,while the induction rate is only 38.33%in normal light.The different time of low temperature treatment has no obvious effects on the induction of anther callus,while the samples without low temperature treatment and the ones with treatment at 4℃for 1 day are both beneficial to induce callus in which the highest induction rate is 80.13%.Time of sample collection also has effects on induction of anther callus,that is,samples should be collected after one week of full flowering stage,and the best ones are the corolla with 2:1 of length and width.2,Varieties and concentration with different ratios of hormones are further studied in redifferentiation of blueberry anther callus.The results show that the best culture medium consists of WPM,0.5mg/L 6-BA and 1.0 mg/L 2,4-D in which the rate of anther proliferation reaches 1.6567g,and 2,4-D plays an important role in the induction of callus.CPPU has the biggest influence on the proliferation amounts of callus than other kinds of Cytokinin with the same concentration,which can reach the highest amounts of callus,however,6-BA and KT,or 6-BA are both more beneficial to the growth of callus.PH value of medium are also studied,and the results show that the culture medium at pH 5.50 has obvious effect on proliferation of callus of blueberry anther in which the proliferation amounts reach 1.7885g,and pH under 5.0 is beneficial to the growth of callus.In proliferation culture study,the best culture medium consists of WPM,1.0mg/L CTK and 0.4mg/L IBA or WPM,0.5 mg/L 6-BA and 0.25mg/L NAA,which are the most beneficial to the proliferation and differentiation of anther callus.3,Polyploid of two blueberry species induced with colchicine are studied and its ploidy are determined.The results show that: Stem Segments Culture."Northland" can be induced into the chimera plants after the treatment by clochicine impregnation and drug culture medium.The stems soaked at 2.0 mg/L for 24h can get the best results of clochicine impregnation,and the rate of induction reaches 32.50%,while 0.002%drug culture medium is the best treatment for the induction of chimera plants with the induction rate of 5%.For the mutated, Colchicine impregnation and drug culture medium can also induce tetraploid cells,but no other ploidy cells can be found that shows the ploidy cells will disappear after repeated culture.Purification.No variation are found after the subculture and selection of the mutated blueberry plantlets,and the chimera plants will be replaced between stratums after the repeated culture,leading to the disappearance of periclinal chimera.
