Research On Asexual Propagation Biology Technique Of Ribes Genus

It has been researched on the asexual propagation of Chinese wild plant Ribes fasciculatum var.chinensis, R.rubrum'Rovada',R.'Jostabeere'and R.rubrum'Jonkheer'which three cultivars were introduced from Germany by different methods of layering and cutting to indicate the effect to the rooting rates and quality of roots by different propagation times,structure growth of twigs,hormone and their contents using the experimental technique of anatomy and ELISA in this thesis.The results shows that:(1)It is easy to root for different species and cultivars of Ribes by the several methods of layering and cutting,and there are some diversity between these species;It needs the most rooting days and has the poorest roots quality for'Jostabeere',and the'Rovada'has rooted earliest and with the best quality in these three cultivars with the mound layering.There are the similar rooting rates and roots quality for three cultivars when using the air layering,and the'Jonkheer'is better than others.(2)It makes the rooting days shorter and improves the rooting rates and roots quality by cutting the layering twigs and treated with hormone,and the best treatments is to cut and treated by 50ppm NAA both in mound and air layering,in the simple layering the best one is cutting and 100ppm NAA.(3)Comparing the rooting result and their environmental factors during cutting time,it has been found that the best season for Ribes's cutting in Beijing are June and September,the suitable temperature of cutting bed is about 20~25℃,and the relative humidity is 96~98%.(4)It can be shortened the inducement and forming stage for roots primordium by NAA and IBA treatment,and NAA is little stronger than IBA for this function,but IBA has a special effect in increasing the roots number and their growth speed and setting up the better root ball system.So it has the best rooting result after treated by 100ppm NAA and IBA,and IBA is much better than NAA.(5)There isn't any latent adventitious root primordium in soft-wood cutting of R.fasciculatum var. chinensis,the type of rooting is the phloem forming adventitious root directly,and the roots originated from the cambium below primary phloem.So it is easy to root for mature of primary structure of twigs in early summer,but it will have difficulty when the secondary structure forming for cutting's rooting.It has been found there isn't a root from callus,though there are callus forming when cutting.It found that the effect of external hormone NAA,IBA and their concentrations have the close relationship with the structure growth of cuttings.(6)When measuring the internal hormone of IAA,ABA,GA and ZR during the twenty-eight days for different five times of cutting from June to September,it is found that the content of the four kinds of internal hormone are much higher in June and September than they are in July to August remarkably,as while the rooting rates and quality are much better than they are in the middle summer.It has been also found that it will have the good rooting result when they are in high content of internal hormone IAA,ABA,GA and ZR on the beginning days of cutting,otherwise,the rooting effect will be worse.And it will be good for the rooting when the changeable of content of IAA,ABA,GA,ZR and IAA/ABA ratio has decrease during the root primordium forming.After the roots growth out,the increasing of their content will lead and promoting to the root growth.(7)Comparing the efficiency of different asexual propagation methods for layering and cutting,it will easily found that it has the fastest growth of roots,shortest period for propagation and higher propagated efficiency for the air layering,and it will have the best root-ball system and strongest young plants with the early maturity plants for mound and simple layering.In these asexual propagation methods,the cutting has the highest efficiency for propagation with a shorter period and stronger plantlets.It is the best way for production and developing of Ribes cultivars in the feature utilization.
