Lily is one of perennial bulbous flowers,which belongs to the Lilium of Liliaceae.Lily is one of the most important cut flowers of the world.China is rich in lily resources and famous for its widespread endemic species.However,the lily resources in China have not been appropriately applicated.In view of this question,the author studied on resource investigation and collection in Sichuan province.Methods for culture in vitro and slow growth conservation of lilies that the author and the research group collected were examined.The cross compatibility of wild lilies and varieties of lily and the interspecific cross in lilium were examined as well.It had important theoretical and practical application value to the preservation and application value of resources of Lilium in China.The main results are showed as the followings:1.The author collected 4 species of wild lily in Sichuan Province.They are L.sargentiae, L.duchartrei,L.davidii and L.regale.The distribution,morphology characteristics,ornamental features, survivorship environment and characteristics of plant community of Lilium species were investigated through sample plot surveys.Both L.regale and L.sargentiae distributed in the areas of low elevation. L.regale had strong drought and heat resistance with strong growth.L.sargentiae grew in the forest edge where was sunny and humid.It had large flowers and grew robustly.L.davidii grew up at the 1700-2500m altitude,in the forest edge and hillside.Its flowers are red and bright.L.duchartrei like cold environment and grew up in the areas of high elevation,where were meadows,forest edges and nearby brooks.The survey discovered the wild lilies were destructed seriously and need to be protected urgently. 2.The lilies collected in Sichuan and by the research group including L.longiflorum,L.davidii var.unicolor,L.lancifolium,'Yelloween','La Mancha','Robina' were used as the materials to tissue culture.The lily scales,bulbils and floral organs were used as explants to establish regeneration system. The results showed that 6-BA1.0-2.0mg/L+NAA 0.1-1.0mg/L+ MS+30g/L Sucrose+6.0g/L Agar was suitable for the most of lilies which had strong differentiation abilities and the medium need to be screened for the ones which had poor differentiation abilities.The bubils were easily propagated and had high differentiation rates.The ovaries and pedicel sections had relatively high differentiation rates when floral organs were used as explants.The research used bulblet leaves for renewable materials to establish an efficient propagation system as well and screened the suitable media:MS+6-BA2.0 mg/L +NAA0.2 mg/L,MS+6-BA2.0 mg/L +NAA1.0 mg/L and 6-BA 1.5mg/L+NAA 0.2mg/L.This research was to lay the foundation for the lily resources conservation.3.The L.davidii,L.longiflorum,L.lancifolium,'Yelloween' and 'La Mancha' were choosed as the materials,and the effects of different temperatures,basal media,sucrose and ABA concentrations on the growth of in vitro lilies were examined.The slow growth system was established.The results showed that the best culture media were 1/4MS +90g/L sucrose and 1/4MS +3.0 mg/L ABA +30g/L sucrose under the -2℃,in which the lily germplasm could preserved well more than 18 months.4.The cross compatibility of wild lilies and varieties of lily,as well the interspecific cross in Lilium were evaluated.The results showed that the degree of relative affinity as the followings:wild species and L.×formolongi>interspecific cross>wild species and Oriental lilies>wild species and Asia lilies.Some hybrids were obtained by embryo rescue techniques,such as the L.regale,Lsargentiae,and L.concolor with L.×formolongi,which provided the basis for the future research of breeding new resistant varieties and genetic regularity of the resistance.
