Mastitis is inflammation of mammary gland because of various causative organism. Mastitis becomes main resource of deleterious bacterium of milk and milk product. It also becomes common with disease of man and animal. Especially , etiological factors of environmental mastitis are very complicating and species of causative organism are relatively much more. Therefore, the experiment carrys out analyses of immunogenicity of main causative organism of environmental mastitis that has been already separated and identified and prepares vaccine. I hope experiment will provide new method and route to prevent and cure environmental Mastitis in Inner Mongolia .To select strains of good immunogenicity and extensive antigen-spectrum, 2 S Escherichia colis and 2 S Klebsiellas are processed with SDS-PAGE and Western-blot . The results show that the protein bands of A1 and K1 are more fortis than A2 and K2. To progress thoroughly immunogenicity analysis, ELISA is established to detect the antibody titers and the cross protective antibody in blood serum of rabbit after immunization. A1 and K1 of typical antigen-spectrum and allround protective antigens are suitable to become strains of preparing vaccine.The bacterial suspension of A1 and K1 is unactivated with formaldehydes and progressed germfree test . Alhydrogel adjuvant is selected to make inactivated vaccine against environmental mastitis and the vaccine is tested with safety. Collecting blood serum of rabbits after three times immunization with the vaccine is tested specific antibody by method of ELISA. The results show the vaccine against environmental mastitis of cows by mixing with alhydrogel is safe. The vaccine also can suscitate rabbits to produce high specific antibody . With the mouse model, challenge test is processed after three times immunization with the vaccine. The results show effective protection against Escherichia coli and Klebsiella of homologous and various serological types could be gained.To estimate effect of prevent and cure bovine environmental mastitis by inoculating inactivate vaccine . To carry out experiment of inoculating vaccine for cows in grazing land of Inner Mongolia Agricultural University. Experiment establishes ELISA to test specific antibody in blood serum and milk serum for evaluating effect of immunity . Testing specific antibody in blood serum and milk serum after three times immunization with the vaccine, the results show that specific antibody of immunity group in blood serum and milk serum remarkably step up than control group and preimmune . Collecting milk samples of cows and determining somatic number, the results show that somatic number of immunity group decrease than control group and preimmune.
