| Lonicera macranthoides Hand.-Mazz.was the plant of Lonicera in caprifoliaceae.It was the local variety of traditional chinese medicine honeysuckle,and cultivated in southwest and central south China.It was one of the chief commodities of the local traditional chinese medicine honeysuckle.It had the effects of heat-clearing,detoxicating,cooling the blood and detumescence. And it was applied to furunculosis,laryngalgia,erysipelas,toxic-heat and blood stasis,wind-heat type common cold and seasonal febrile disease.In the Chinese Pharmacopoeia 2005,it was entered in the new varieties flos lonicerae,as the original plant of flos lonicerae.A systematic research on the vegetative propagation of Lonicera macranthoides Hand.-Mazz.had not seen.This paper had carried on cutting techniques of Lonicera macranthoides Hand.-Mazz.and its rooting mechanism for further research,which can provide theoretical basis for the study of cutting propagation.The research redults as follows:It was found that the exogenous hormone can increase the rooting efficiency.The rooting efficiency of the cuttings treated by ABT6 was better than by NAA and IBA.The rooting position of cuttings treated by exogenous hormone had greatly changed.The number of cuttings in node and intemode was much more than in incition.The study of the effect of different concentrations of exogenous hormone in the cuttings showed that the optimum concentration of NAA,ABT6,IBA was respectively 700mg·L-1,500mg·L-1 and 300mg·L-1.In certain extent,the higher the concentration,the better the rooting efficiency.Higher than the optimum concentration,the rooting efficiency will be decreased.The optimum soaking time was respectively 700mg·L-1 NAA30s, 500mg·L-1IBA10s and 300mg·L-1,ABT660min.the best cutting medium was muck:pedite=1:1.The ralationship between nutrient dynamic change and rooting was as follows:the trend of the soluble sugar and starch in leaves and basel of cuttings was first decreased then increased,the occurrence period of adventitious root was the lowest,positive correlated with rooting rate, (r=0.775,0.700,0.139,0.274).The Soluble Protein in leaves was also first decreased then increased,and in basel of cuttings,it showed downtrend,at later stage,it tended to mitigation. positive correlated with rooting rate(r=0.515,0.593).The total nitrogen content in leaves and basel was downtrend and later mitigated.They had negative correlation with rooting rate(r=-0.422, -0.500).The C/N ratio in leaves and basel of cuttings was first decreased then increased,and later mitigated.They had positive correlation with rooting rate(r=-0.771,0.668).The trend of total phosphorus in leaves was firstly decreases and then increases,and in basel of cuttings,it was gradually decreased.They had positive correlation with rooting rate(r=0.569,0.655).The trend of chlorophyll content in the process of rooting was that in the adventitious root primordium induction period increased,decreased in formation period and in elongation period increased.It had positive correlation with rooting rate(r=0.625).The trend of tissue water content and relative water content in leaves and basel of cuttings was first decreased then increased.The valley value appeared in formation period of adventitious root. positive correlated with rooting rate,(r=0.525,0.874,0.793 and 0.366).The ralationship between rooting and protective enzyme activity in leaves and basel epidermis was as follows:the MDA contents first increased,and decreased in adventitious root's formation period,negative correlated with rooting rate(r=-0.323,-0.499).SOD activity in leaves decreased in the frist 12 days,then in formation period it increased.And in basel epidermis of cuttings it was first increased then decreased.SOD activity had positive correlation with rooting rate(r=0.449, 0.757).POD activity was also first increased then decreased,positive correlated with rooting rate (r=0.631,0.732).CAT activity in leaves kept reducing,while in basel epidermis,it was first decreased then increased,negative correlated with rooting rate(r=-0.665,-0.596).The ralationship between rooting and endogenous hormones in leaves and basel epidermis was as follows:the IAA contents first decreased then increased and later decreased last mitigated, positive correlated with rooting rate(r=0.802,0.860).ABA contents in leaves first increasted later mitigated.And in basel epidermis,it first increased then decreased later increased last mitigated, negative correlated with rooting rate(r=-0.388,-0.778).The trend of IAA/ABA was similar to IAA contents,positive correlated with rooting rate(r=0.352,0.937).GA3 contents in leaves decreased in the frist four days,and then mitigated,negative correlated with rooting rate(r=-0.499).In basel epidermis it changed with a larger fluctuation,positive correlated with rooting rate(r=0.887).The anatomical study on the rooting of Lonicera macranthoides Hand.-Mazz.'s cuttings showed that it belonged to non-callus rooting,and it was induced root primordia. |
