To investigate the nutrition metabolism of Agrocybe chaxingu Huang polyphenol and antioxidation activity of its metabolites, we used Camellia oleifera Abel branches, the host tree of Agrocybe chaxingu Huang in nature, as the nutrient additive for liquid culturing, and preliminary studied the effects of polyphenol nutrition metabolism induced by Camellia oleifera Abel branches. By analyzing the nutrient components of powder of Camellia oleifera Abel branches and its antioxidation activity (step 1), we investigated the feasibility of using it as base material in the liquid culture of Agrocybe chaxingu Huang and the range of adding volume. By comparing the differences of biomass production ,total phenol content of the metabolites, total sugar content and antioxidation activity using different adding volume (step 2), we determined the suitable adding volume. Morover, we compared the nutrient components with the suitable adding volume and the characters of the nutrition metabolism of total phenol, total sugar and total nitrogen (Step 3).The aim of this research is to determine the possibility of using Camellia oleifera Abel branches as the culture base material, and thus to increase the content of antioxidation material in the metabolism of Agrocybe chaxingu Huang and to improve its antioxidation activity. The results of step 1 indicated that using Camellia oleifera Abel branches as the culture base material is able to promote the nutrition metabolism of antioxidation components. The total phenol content of the extracts from Camellia oleifera Abel branches was 7.12mg/g. When total phenol content of the extracts was 79.13ug/mL; the inhibition rate was 53.14% in egg yolk lipoprotein fatty acid (PUFA) system, and 11.17% for superoxide anion radical in Pyrogallic acid system.Comparing the adding level of 2%, 4%, & 8% of Camellia oleifera Abel branches into culture medium, it showed that adding level of 8% significantly raised the biomass of mycelium of Agrocybe chaxingu Huang(1.93g/100mL,p<0.05). For both of hot water extracts and extracellular metabolites, the total phenol of 8% group was higher than CK, 2%, and 4% group. The inhibition rate of oxidation in PUFA system for 8% group was higher than CK (P<0.05). The conclusion was that the total phenol content and the inhibition activity of both hot water extracts and extracellular metabolites were improved by adding 8% branches of Camellia oleifera Abel into culture medium. The dynamic analysis results of nutrition metabolism of 8% group showed that the total phenol content in liquid culture was increasing with the prolonging of culture period, and the total phenol content in liquid culture in 8% group was obviously higher than CK group, but the total nitrogen content was decreasing gradually, and total sugar content had small changes and reached to stability. According to nutrition condition of culture medium, the culture period of culture medium, into which added 8% Camellia oleifera Abel branches, should not be longer than 5-7 days. Moreover, it is important to carry out further research on its intermediate product and dynamic accumulation of phenol metabolism of Agrocybe chaxingu Huang.
