| The major factors affected the Lamb superovulation-Ovum Pick-up(OPU)and in vitro fertilization (IVF) technique were discussed systematically in this study in order to establish an acceptable production system of Lamb goat super-ovulation-OPU-IVF in our lab. The objective of the present study was to improving the in vitro developmental capacity of lamb goat embryo obtained by superovulation and OPU.Development of induced follicular. Lambs were divided into three groups by different FSH manufacturers. The result indicates that, in oocytes amount average per lamb, the difference between Japanese FSH group and Native (NingBo) FSH group was significant (66.0 vs 21.0, P<0.01). And the average oocytes amount in Japanese FSH group show no significant difference with New Zealand FSH group (66.0 vs 55.75, P>0.05). Average oocytes available for fertilization in imports FSH groups (25.5 & 31.75) were significantly (P<0.05) higher than native FSH group (7.75). Comparison of the effects of day-old on oocytes amount obtained after superovulation and OPU. Average oocytes amount and average oocytes available for fertilization (64.67 & 31.67) in 30~50 day-old group were very significantly higher than 51~70 day-old group (31.0 & 11.67, P<0.01). One month later, superovulation was repeated on 4 lambs.The data shows that oocytes obtained after the first treatment was more than the second time (43.50 vs 8.75). On the contrary, the cleavage rate in the first batch oocytes was much lower than the second time (58.25% vs 13.5%).The results of research on lamb goat IVF efficiency and developing capacity show that, several factors, such as co-incubate time in IVF medium and different pH of IVF medium, influence the cleavage rate and developing capacity of oocyte. Oocytes were divided into three groups by different co-incubate time in IVF medium. Cleavage rate in 23~26h (48.33%) group was very significantly (P<0.01) higher than other two groups (16.0% and 21.33%). When IVF medium pH was at the level of 7.5~7.6, cleavage rate (51.8%) was much (P<0.01) higher than pH=7.7~7.8 group (29.8%). The difference of morula rate between the two groups was not significant (33.3% vs 27.3%, P>0.05).50 embryos obtained from our IVP (in vitro embryo production) system were trans- fered to 12 receptors. Three of them were pregnant which tested by B-ultrasound device 40 days later. Pregnancy rate was 25.0%, five lambs were born by two receptors at last. Then blood of new lambs, donors and receptors was collected to analysis their DNA sample, further, to prove that the new lamb's DNA comes from the donor's DNA.Compare the IVF efficiency and development capacity between adult and lamb goats. Cleage rate of lamb group was significantly lower than adult group (34.9% vs 45.9%, P<0.05), and the morula rate continue with the same trend (17.24% vs 35.9%, P<0.05). Blastocyst rate in adult group was higher than in lamb group (23.9% vs 10.7%, P<0.05). |
