Self cross future generation (T1 generation) of p5CS transgenic wheatgrass were used as material in this study.By molecular and resistence determination,stably transferred wheatgrass were obtained and the integration and expression of exogenous gene p5CS in T0 and T1-generation transgenic wheatgrass were confirmed,which provide theoretical basis for the later study of transgenic wheatgrasses;at the same time, transgenic wheatgrass with strong resistance were identified by detecting there drought and saline tolerance,as basic materials for breeding high tolerance varieties next step.The preliminary findings of this study are as follows:1.The exogenous gene p5CS have integrated into the genomic DNA of T1-generation transgenic wheatgrass and the integration number is 1~2,which confirmed by PCR and Southern blot analysis.Northern blot analysis showed that some of the exogenous gene in the T1-generation transgenic wheatgrass expressed at transcriptional level.There are 58 plants presented positive result in a total of 128 T1 generation transgenic wheatgrass in PCR analysis.2.Under drought stress and salt stress,the change of chlorophyll content and soluble sugar content of T0 and T1- generation transgenic wheatgrass showed no difference compared with non- transgenic wheatgrass.On the 14th day of intimidation,the enzyme active of SOD,POD,CAT in transgenic wheatgrass were a little higher than non- transgenic wheatgrass.The Leaf relative water content and Pro content of transgenic wheatgrass were higher than non- transgenic wheatgrass;The membrane permeability and MDA content of transgenic wheatgrass increased less than non- transgenic wheatgrass.Taking all these results,the tolerance of transgenic wheatgrass were better than non- transgenic wheatgrass.3.The Pro content of T0 and T1- generation transgenic wheatgrass can Increased 2-fold of comparison plants,that proved the exogenous gene p5CS have stably transferred into T0- generation transgenic wheatgrass,and have passed into T1- generation transgenic wheatgrass,and expressed.
