Euphorbiae bracteolata Hayata,Daphne euphorbia fisheriana and Euphorbia fischriana are the main plants for Euphorbia fischriana medicine.They have many kinds of biological activities like anti-tumor,anti-virus and bacteriostasis insect disinfestation.But the research maily focus on Daphne euphorbia fisheriana,the chemical component and its effect of Euphorbiae bracteolata Hayata is less be researched.Fusarium Wilt of Tomato is one kind of disease severely influence the quality and output of vegetable that caused by Fusarium oxysporum Schl,it is mainly controlled by chemical pesticide.Pesticide that produced from plant is uncontaminated and no-residual,so it is an effective way to solve the pesticide residues problem by use this kind of pesticide to displace the chemical pesticide.For those problems,this study used Euphorbiae bracteolata Hayata as raw material,total flavones was extracted and purified from its root.Then the inhibiting effect by the total Flavones to Fusarium oxysporum Schl in vitro and the control or prevention effect by the total Flavones to tomato seedlings infected by Fusarium Wilt was been studied;On this base,the activity of a series of enzyme related with disease resistance was researched,when the tomato seedlings were dealed with the total Flavones from Euphorbiae bracteolata Hayata.This study can provide theoretical basis for the exploitation and utilization of Euphorbiae bracteolata Hayata and biological prevention to plant disease.The main research are as follows:1.The optimal conditions on total flavones extracted of Euphorbiae bracteolata Hayata.were as follows:time of extracting at 2h,temperature of extracting at 70℃, ratio of material to fluid at 1:40 and concentration of ethanol at 50%.The order on purification is ratio of material to fluid>time of extracting>concentration of ethanol>temperature of extracting and the significantly factors are ratio of material to fluid and time of extracting.2.The optimal conditions on total flavones purified of Euphorbiae bracteolata Hayata.were as follows:concentration of adding liquid at 20mg/L,pH of adding liquid pH at 4.0,concentration of ethanol at 50%and quantity of adding at 1ml/g.The order on purification is concentration of adding liquid>pH of adding liquid>concentration of ethanol>quantity of adding and the significantly factors are concentration of adding liquid and pH of adding liquid.3.Purification of total flavones on the roots of Euphorbiae bracteolata Hayata. had significantly inhibitive effect on the mycelia growth and spore germination of Fusarium oxysporum.The rate of inhibition rised with the increasing of concentration of total flavones,and when the concentration reached at 20mg/L the rate inhibition was 100%.The mycelia treated became thinner,ramus and transparence decreased, something thick assembled,more vacuole appeared and became bigger.The penetration of membrane increased and the activity of SOD,CAT increased first and decreased later.4.Disease index on tomato seedlings after treatmenting 7d and 14d by extracts and purification of total flavones on the roots of Euphorbiae bracteolata Hayata. increased with total flavones contention decreased.Extracts concentration is at 40mg/L,disease index is at 24.85%until 7d and at 50.17%until 14d.Purification concentration is at 30mg/L,disease index is at 20.98%until 7d and at 46.87%until 14d.Disease index of purification on Fusarium oxysporum.is batter than extracts.5.Preventive effect of tomato seedlings on total flavones extracted and purified from the roots of Euphorbiae bracteolata Hayata.show that when extracts concentration is at 15mg/L,preventive effect is at 48.39%until 7d and at 25.18% until 14d.Purification concentration is at 15mg/L,preventive effect is at 23.36%until 7d and at 12.72%until 14d.Preventive effect of extracts on Fusarium oxysporum.is batter than purification.6.After tomato seedlings were treatmented by extracts and purification of total flavones on the roots of Euphorbiae bracteolata Hayata.,the activity of SOD,POD, PAL,PPO,β-1,3-glucanase,chitinase and lignin content increased.SOD and PAL are the key enzymes on tomato seedlings resistance inducted and preventing Fusarium oxysporum.when tomato seedlings t were treatmented by extracts and purification of total flavones on the roots of Euphorbiae bracteolata Hayata.
