Plant surface is covered by a layer of epicuticular wax. It can regulate non-stomatal water loss,protect against ultraviolet radiation damage and resist the invasion of germs. Studies have showed that a positive relationship between the content of epicuticle wax and plant stress resistance. Characterization of gene functions according to the changed characters of overexpression and RNAi silenced or reduced expression transgenic plants has be an effective methods in recent years.Spatial expression pattern of the three rice WAX2 homologs(OsWAX2-1, OsWAX2-2 and OsWAX2-3) were analyzed using semi-quantitative RT-PCR. The result showed that the three genes were expressioned differently. There were also differences in expression patterns among the three OsWAX2 genes when treated on high-temperature, low temperature, NaCI, PEG and ABA. These results provided reference for identifying the functions of these three OsWAX2 genes.Agrobacterium tumefaciens-mediated transformation method was used to transfer OsWAX2 gene overexpression vectors and RNAi vectors into rice. The explants, media and culture conditions have been optimized for the transformation. Gene transformations were conformed at DNA level for both overexpression and RNAi transgenic plants. Gene overexpression was detected in overexpression transgenic plants, but inhibited gene expressions were not detected in RNAi transgenic plants. Phenotypes of OsWAX2-1 overexpression and RNAi T1 transgenic plants. The research indicated that OsWAX2 may be involved in the epicuticular wax synthesis and play a positive role in adversity resistance. Further research should be conducted to identify the function of OsWAX2.
