Study On The Technique System About Tissure Cultrue And Rapid Propagation Of Dendrobium Nobil

Dendrobium NobiI is the crossbreed of Chese Dendrobium nobil Lindl.It blossoms always in spring and is used as pottesd plant.Becuase of its manifold color of flower and beautiful gesture,it is popular with European and Japanese.But now because over all level of professional production of Dendrobium Nobil is not high,and the key technologies of production are weak,those result in higher production costs,low propagation coefficient and inefficient industrial production.In the same time,many expensive varieties are in shortage,low quality and high market prices.Therefore Dendrobium Nobil tissue culture research is the key to solving this problem.The axiUary buds of the false bulb of Dendrobium Nobil were taken as explants to induce bud clumps and this regeneration way of propagation were studied in this experiment.This experiment resolved around the best ways to exterminate germs of explants,producing cluster buds,proliferating cluster buds,strengthening test-tube seedlings,engending roots and transplanting finally.The main contents focus on the following aspects:the selection of explants,asepsis methods,basic culture mediums and raising modes,the combination and density of hormone,different addictions,carbon source.The Orthogonal Design is adopted to arrange experiments,and all data analysis was dealt with by SPSS.The result shows that the effective procedure of setting up asepsis shoots is follow: the axillary buds of the Dendrobium Nobil caudexes→washed by tap water→dipped in the detergent water about 10min and used a soft brush to clean the surface dirt→washed about 30min by tap water→putted on the extra-clean desk→dipped in the 70%alcohol about 20s→washed 4times by asepsis water→dipped in the 0.1%HgCl₂15min→washed 8 times by asepsis water→water dried and the bracteal leaves sweeped away→inoculated.The significant factors of inducing cluster buds is the basic culture medium,BA and natural additions,the subordination factor is NAA.The best inducement culture medium is 1/2MS+BA1.5mg/L+NAA1.0mg/L+CM15%+sugar30.0g/L+ agar7.0g/L,pH5.4.The main factors of buds multiplication are the basic culture medium,KT,BA, NAA,and the pH of culture medium,the height of seedling,the density and mode of seedling,and natural additions.The pH5.4 is best.The seedlings about 1.5cm cut vertically at the bottom can obviously improve the multiplication of bud clumps.The best density is 4 seedlings/bottle and banana is the suitable additions.The best multiplication culture medium is 1/2 MS+BA 1.0mg/L+NAA 1.5mg/L+KT 0.5mg/L +sugar 30.0g/L+agar 6.0g/L+banana juice100.0g/L,pH5.4.The best culture medium for promoting seedlings and radication is meliorative MS +BA 0.5mg/L+NAA1.0mg/L+banana juice 100.0g/L+sugar 30.0g/L+agar 7.0g/L, pH 5.4.When transplanting,first,seedlings are forged in the room for 7days,then transplanted.Sphagna is the best soilless media for Dendrobium Nobil,the living rate can reach 91.67%,meanwhile,seedlings grow strongly,and leaves glitter and all of them are upstanding.Through the research above,the main effected factors and the suitable culture medium in each stage were studied fully,then the technique parameter combination were filtrated,the superior regeneration system about Dendrobium Nobil to expand the choice scope of explants,improve the propagation coefficient,and,thus enhancing Dendrobium Nobil commercial production efficiency,prov,de solution to the current market shortage of fine breed Dendrobium Nobil,and technical support for professional nursery.
