A Preliminary Study On Isolation And Physiological Characteristics Of Methanogenic Bacteria

Methanogens is a special prokaryotic organism which produce large quantities ofbio-gas as the end mumber that exist in various extremist anaerobic environments to produce energy.As the final member of anaerobic biological treatment in natural carbon cycle,it transform large number of organic matter into renewable energy with other flora bacterias synergy.It play important role in promoting the cycle of the nature material and solution for the energy crisis..Methanogens is the core of methane fermentation,development the bio-gas which is the key to solution the fural energy,So the contents of menthanogens in the firedamp pool and the growth directly impact on the quality of gas.This study simply summaried the biomass and biogas fermentation which closely related with methanogens, and the methanogens' role in methane fermentation(anaerobic digestion),probably summaried it's physiological characteristics and ecological characteristics,it's research progress and research significance.According to the methanogens's metabolism and the growth characteristics in the environment,the study optimize the medium and the specific separation methods,obtained the medium and the separation methods which are suitable for methanogens's growth.The culture medium with a mixture of chemicals was tested for the isolation of methanogenic bacteria.About the methods,The first time to use the small inverted glass to collect the gas which is processed by methanogens,and as the basis to determine whether there is any methanogen growth,the first time to analyse the metabolites ingredients.This study obtained five pure methanogenic bacterias by used the optimal medium and separation method, according to their morphological and biochemical characteristics,Initially identified as the acid methane bacilli sp. And first to analyse the composition of the metabolites,obtained methanogenic bacteria's content of the methane which is more than 98%.According to the growth of it's feature,this study reported the methanogenic bacteria ' s preservation methods.In this study,five methanogens' growth matrix,PH value,temperature,tolerance the salt concentration and penicillin concentration ware studied,five string methanohens are used only acid sodium for grouth,they optimum growth range of PH value are 6.0to8.0,The optimum growth temperature is 37℃,the tolerance to the largest salt concentration of 2%to4%,And the most resistant to penicillin concentration is 1.28to2.56 million units / ml range.
