Analysis On HMW-GSs Of Wheat Germplasms And Selecting Wheat New Lines Using HMW-GS Markers Assisted

Based on characterization of the agriculture trait and combinations of high molecular weight glutenin subunits(HMW-GS) of 182 wheat materials,four with integrated agriculture trait and high quality HWM-GS were selected to polymerize cross-fertilize Using HMW-GS as markers assisted to select new lines in the F2 separate generation,especially the lines polymerizing HWM-GSs In the after generations,traits related to output were charactered firstly,marker assisted selection using HMW-GS markers for high quality wheat lines were studyed The main results were as follows:1.In the analyzed 182 wheat materials,three kind of HWM-GS in Glu-A1 were found,which were Null,1 and 2*.Among them,the frequency of Null was the highest(58.80%).There were ten kind combinations of HMW-GS in Glu-B1,which were 7+8,7+9,14+15,6+8,8,7,17+18,13+16,6 and 21 Among them,the frequency of 7+8 and 7+9 were 38.47%and 34.62% respectively,which was more higher than the other types.Six kind combinations of HMW-GS in Glu-D1 were found,which were 2+12,5+10, 5+12,4+12,2+10 and 10.Interestingly,2+12 appeared most frequently,which accounts for 70.33%41 kinds of HMW-GS combination of diffrent types were examined Among them,"N,7+8,2+12","N,7+9,2+12","1,7+8,2+12" and "1,7+9,2+12" appeared frequently,the frequencies were 20.33%,18.69%, 9.34%and 7.70%respectively Some combinations of subunits with high quality were also examined.In analysis of the home-land wheat varieties,the subunits 5+10 and 2* were less But the subunits 5+10 and 2* appeared more frequently in the foreign wheat materials which also had some rare types of subunit combinations.2.SDS-PAGE analysis of HMW-GS in four F1 positive and negative cross-fertilize materials were conducted.The offspring all had the same bands as their parents This accorded with codomiance.The contents of expressed HMW-GS leaned to female Analysis on HMW-GS in four F2 separate colony showed that the ratio of Glu-A1a(1) to Glu-A1c(N) in accordance with certain rules(1:1) in the Dlu-A1 locus,the ratio of Glu-B1f(13+16) to Glu-B1b(7+8) and Glu-B1i(17+18) to Glu-B1f(13+16) and Glu-B1f(13+16) to Glu-B1h (14+15) and Glu-B1h(14+15) to Glu-B1i(17+18) in accordance with certain Rules(9:7) not the law separation of allele(1:1) in the Dlu-B1 locus Phenomenon of silence of X subunit or Y subunit in different loci were found.3.Analysis of the relation between different HMW-GSs and wheat quality showed that different subunits from the same loci had the different effect on the wheat main quality,main quality character of different lines from same cross-fertilize combination were different and some lines exceeded their parents These results showed that different subunits or subunit combinations had different effect on wheat quality character,the genetic effect and genetic background of HMW-GS to quality character had effect one another High quality subunits or high quality subunits combinations could improve and enhance main quality characteristic of wheat.Using HMW-GS markers assisted to select wheat quality character was an effective method.4.14 wheat new lines polymerizing high quality HWM-GS were selected based on HMW-GS marker assisted selection in the F2 separate generation,especially selecting high quality HMW-GS polymers and charactering traits related to output in the after generations.Among them,5 new lines not only were high quality HWM-GS polymer,but also provided with high quality character and resisted mostly manifold disease They had colligated excellent agriculture trait These polymer of high quality HWM-GS could be used not only as parents in cross-fertilize breeding but also in agriculture manufacture after further output experimentation.
