| Great achievements have been made in using and studying rapeseed hybrid vigour in China, and China has done significant contribution to the international rapeseed studying and utilization. For example, Fu find the first valuble rapeseed cytoplasmic male sterility(pol CMS) in the world wide. After that, Song et al indicate the DGMS under genetic hypothesis of one gene with multiple, and Hong find Rs1046AB Mf is allelic to Ms. 20 sterile plants from F2 progenies generated by crossing restorer 195A-14 with Rs1046A were randomly selected to test crossing with maintainer 7-5, and no fertile individual was observed in test cross F1, indicating Mf from 195A-14 is allelic to Ms from Rs1046A, which confirmed the multiple allelism hypothesis again. He also has designed markers linked with the restoring gene. The marker-assisted selection technique has been successfully used in rape cross breeding, and the MAS make the foreground selection fast with less linkage drags. At the same time, background selection has also been much more efficient in this way. Generally speaking, the MAS is a good way to select restor line.Our research taked the 195A (pol CMS) as the donor of restoring gene and taked the 6275 as recurrent parent to select restor line with the marker-assisted selection technique. In this research, we did the foreground selection and the background selection in BC1 progenies and BC2F1 progenies. We used the SCAR markers SCD7-1,FR2L2 in the foreground selection, and we got the same result. After that, We got 64 target individuals from 131 individuals in BC1 progenies and 93 individuals from the 258 individuals in BC2F1 progenies. Then, we did the background selection with the AFLP technique and got the The UPGMA cluster map with the software NTSYS2.0. Finally, we got the individual which had the best background.The restor line we got have been one of the excellent germplasm resourcese for the future breeding. |
