Ferula sinkiangensis K.M.Shen, a kind of useful Chinese peculiar medicinal plant and at the edge of extinction,was selected as the research material. The seeds dormancy mechanism and rapidly propagation of Ferula sinkiangensis were researched in this paper. The propagation system was preliminary established and it provided a feasible approach for protecting and developing the resources of Ferula sinkiangensis K.M.Shen. The main results are as follows:1.The permeability of water and air of Ferula sinkiangensis seed coat was better and it had no effect on seed dormancy. The developments of embryo were completely, when the seeds matured. The seed had some growth inhibitors.2.The influence of the illumination was not prominent for germination. The best temperature for germination was 4℃to 10℃.Stratification in low temperature(4℃) for 40 days was the optimal approach for germination. Stratification in changing temperature for 50 days could break the seeds dormancy. The best GA treatment was 1000 mg/L. After 6 days treatment, the seeds could germinate. The final germination rate arrived 22.6 percent.3. 75%ethanol 10s +0.1% HgCl2 15min +1/2MS medium and stratification in low temperature(4℃) was the best method for attaining no vaccine of Ferula sinkiangensis. MS+0.5mg/L GA could break embryo dormancy. The cotyledon of Ferula sinkiangensis in 1/2MS medium, which contained 1mg/L 2,4-D + 0.1mg/L6-BA +10g/L PVP, could induce callus.The hypocotyl of Ferula sinkiangensis in MS medium,which contained 0.5mg/L2,4-D+0.5mg/L 6-BA+10g/LPVP ,could induce callus.4.Cultivation in greenhouse, the seedlings of Ferula sinkiangensis should be given humid soil, high humidity air and fitting light. The fiting soil layer thickness was 2cm and the transparency was 70 percent. |
