Soybean is an important economical crop. Soybean is the major plant origin of protein and oil for people consumption, and material of feeding stuff. Now, the varieties of high oil soybean and high protein soybean, can not afford the need of people's life and production. So it is useful to use modern breeding technology to modify the content of oil and protein in soybean. Oil and protein are quantitative traits which were controlled by many genes. It has important meaning for breeding application to locate these quantitative traits in soybean linkage groups.The population of 154 recombination inbred lines derived from the cross of America cultivar Charleston and Chinese cultivar Dongnong 594. Oil and protein content of soybean in five different sites were analyzed during 2002 to 2006 year. QTL that underlying oil and protein content of soybean were found.1. 34 QTL were found has relation with soybean oil content in ten environments. Located on A1,A2,B1,B2,C2,D1a,D1b,F,G,H,L and N linkage groups, five QTL were located on B1 linkage group, four QTL were located on A1,D1a and F linkage groups. OilF-3,OilN-1 and OilN-2 were main effect QTL of soybean oil content. OilF-3 were located in the interval of GMRUBP-Satt030 on F linkage group, OilN-1and OilN-2 were located in the interval of Satt022-GMABAB and GMABAB-Sat095 on N linkage group, respectively.2. 37 QTL were found has relation with soybean protein content. Located on A1,B1,B2,C2,D1a,D1b,E,G,J,L,N and O linkage groups. Seven QTL were located on C2 linkage group, six QTL were found on D1a linkage group, and five QTL were found on E linkage group. ProC2-4 and ProE-4 were main effect QTL underlying soybean protein content. ProC2-4 was located on C2 (Sat092-Satt460) linkage group. ProE-4 was located on E (Satt452-Satt231) linkage group.3. In this research, 19 pairs epistatic QTL were found that has relation with soybean oil content, located on eighteen different linkage groups, all soybean 20 linkage groups had QTL was located except C1 and K linkage groups. 20 pairs epistatic QTL were found they has relation with soybean protein content, distributed on thirteen different linkage groups. Most linkage groups had QTL underlying protein content. No epistatic, was found on the same linkage group, had relation with each othert at the same time. But many epistatic QTL, found on different linkage groups, had relation with each other at the same time. It identified the universality and complexity.4. 34 QTL underlying oil content, were located in ten environments. 23 QTL were located in one environment. 37 QTL underlying oil content, were located in ten environments. 27 QTL were located in one environment. So it can be concluded that the expression of QTL underlying oil differed from QTL underlying protein. And QTL underlying oil and protein were affected by environment.
