Activator Protein is a kind of newly discovered proteinaceous elicitor isolated from fungi. Previously studies have demonstrated that it is not only able to increase plant growth but also could induce resistance to pathogen. It is one new type of "green" biological fungicide, with high efficiency, wide resistance spectrum and no residue in the environment. This article studies the culture of the Magnaporthe griesea and the purification of the protein and effect of the Activator Protein from Magnaporthe grisea on Plant Growth and Physiological Activities. The influence and mechanism of activator protein with different concentration on physiological characteristics of Soybean was studied. It showed that the activator protein plays important roles in plant growth and stress tolerance after application. It is useful for further business manufacture.1,The culture of the Magnaporthe griesea and the purification of the proteinThe Magnaporthe griesea was cultured in the liquid PDA medium, and then abstracted the total proteins by Tris buffer. The total proteins were concentrated by ammonium sulphate fractionation, cold alcohol precipitation, and then purified a certain activator protein by anion exchange chromatography and size exclusion chromatography. The result showed that the protein can be purified by the MES buffer at the pH7. 5. The elution of the single peak after size exclusion chromatography showed a single band on the SDS-PAGE.The pI of the purified Plant activator protein was acquired by 2-DE, after analyzed the 2-DE result, we got the protein' s pI about 4. 314±0.035.2,In order to approach the mechanism of enhancing plant growth by new fungal activator protein, the influence of activator protein with different concentration on physiological characteristics of many Crops was studied. The results showed that the seeds of cotton could significantly promote the germination vigor and germination rate of seed after soaking in 2-3μg/mL new type activator protein for 6 hours and the seeds of tomato and loofah soaking in 1μg/mL new type activator protein for 6 hours could significantly promote the germination rate of seed. The seeds of cucumber soaking in a concetration of 0. 2%-0.1% of new type activator protein could significantly promote the germination rate of seed. In order to approach the mechanism of enhancing crop of field growth by new fungal activator protein, the influence of activator protein with different concentration on physiological characteristics of nectarine,garlic,Chinese date was studied. the testing results showed that in 1000-1500 new type activator protein crop of field will growth well.3,In order to approach the mechanism of enhancing plant growth by new fungal activator protein, the influence of activator protein with different concentration on physiological characteristics of Soybean was studied. Compared with control after7d treatment with 1, 2 and 5μg/mL activator protein, root activity of Soybean was increased by 44. 2%,54. 5% andl9. 8%.4,The activator protein from Magnaporthe grisea could ignificantly promote seed germination and young seedling growth The activities of cellulose and the contents of hydrogenperoxide and praline of plant were enhance d after treatment, which play important roles in promoting plant growth and improving stress tolerance.
