In this paper,we made an intensive study about traditional tissue technology and photoautotrophic micropropagation system designed by ourselves of Acacia podalyriifolia,used orthogonal design,through the research effect of different pretreatment ways,subculture medium,sturdy medium,radication medium,and photoautotrophic micropropagation condition and nutrient element on seed sprout and rootage of A.podalyriifolia,and compared the difference culture ways between physiological characteristic and transplant survival on infant A.podalyriifolia, screened out preferable medium and culture condition,explored the new method of the regeneration system building.The main results from our researches were:1.Different pretreatment ways had different effect on the seed germination rate of A.podalyriifolia,No3 indicated that the best way occurred by the pretreatment of thick vitriol for 25 min with boiled water for 20 min,and the germination rate reached 98%.2.Different subculture medium had significant effect on multiplication modulus of A.podalyriifolia,M9 was the best disposal that multiplication modulus reached 3.11. The research result indicated that the best subculture medium of A.podalyriifolia was MS+BA(0.8mg/L)+IBA(0.8 mg/L)+KT(0.2 mg/L).3.Different sturdy medium had significant effect on seedlings increment of A. podalyriifolia,M13 was better than other,reached 31.387 mg.The result was as followed that the proper sturdy medium of A.podalyriifolia was MS(improved)+IAA(0.4 mg/L)+IBA(0.2mg/L).4.The sturdy seedlings had different appearance because of different radication medium on A.podalyriifolia,M27,of the fit radication medium,had the highest radication rate of 81.8%occurred by 1/4MS+ NAA(0.2mg/L)+IBA(0.8mg/L).5.The effect of rooting were different in distinct photoautotrophic micropropagation condition of circumstance about A.podalyriifolia.As a whole,the photoautotrophic micropropagation condition treatment G4,G7,G11 were better than others,in which G4 was suitable for the inducement of rootage,the rooting rate of A. podalyriifolia were 87.5%;The better culture condition of podalyriifolia photoautotrophic micropropagation rootage inducement were:basic culture medium (sand:vermiculite=3:1)condition,dark culture at 0-3 d,4-8 d low illumination culture(2000-3000 Lx),illumination time 10 h/d;later using strong illumination (6000-7000 Lx),illumination time 14 h/d;0-7 d control the extension of CO2 content with 1000-1500 mg/kg,CO2 circulation time 8 h/d:8-12 d control the extension of CO2 content with 1500-2000 mg/kg,CO2 circulation time 12 h/d:after that control the extension of CO2 content with 2000-2500 mg/kg,CO2 circulation time 14 h/d;this was a better culture condition and was suitable for A.podalyriifolia photoautotrophic micropropagation rootage inducement.6.In the better process of photoautotrophic micropropagation,different nutrient element had different effect on the rate of rooting and grade criterion of G22,G24 were higher than other.In all,the water culture medium,including IBA800 mg/L and NAA200 mg/L,were well and the rate of rooting of A.podalyriifolia could reach 91%.7.Compared with the physiological characteristic of rooted seedlings in the different mode of photoautotrophic micropropagation and traditional tissue culture, the research found that the best radication medium of infant A.podalyriifolia in photoautotrophic micropropagation system.The biomess of plantlet over and under Soil,the chlorophyll content,the root vigour were higher than other about 9.44%, 11.38%,55.5%,44.65%.8.Comparing the transplant survival rate of rooting between photoautotrophic micropropagation and traditional tissue culture conditions in different transplantation media,our found that survival rate was higher in the mode of photoautotrophic micropropagation.In two mode,transplantation survival rate of all seedlings had the same change current in four mediums,sand culture medium was the best one and transplantation survival rate reached 89.67%and 78.07%in photoautotrophic micropropagation system and traditional tissue culture.9.In the contrast of photoautotrophic micropropagation mode and traditional tissue culture,they had great differentia in culture cost,the first one could reduce cost notably,the cost of one seedling of photoautotrophic micropropagation mode reduced about 34.18%.
