Study On The Mechanism Of "QiLing" Medicament On Immunodepressed Mice

The immunodepressed mice model were built by intraperitoneal injection with 75mg/kg body weight cyclophosphamide(CY)to normal mice,and the immunodepressed mice were gastrogavage administered with different doses QiLing medicament(doses of QiLing medicament werel2.5g/kg.BW,25g/kg.BW,50g/kg.BW).Firstly,the condition of the proliferation of splenic.lymphocyte,IL-2mRNA levels in T cells,antioxidation index of serum and spleen tissue,and immunity index of serum and protein contents in serum and the activity of ALT,AST in serum were detected.Then,the microstructure and ultrastructure changes of spleen tissue were observed.Through all the experiments above,the effects of QiLing medicament on immunodepressed mice were studied systematically.The results were as follows:(1)QiLing medicament could stimulate the proliferation of splenic lymphocyte,the middle dose group and high dose group were notably different to CY model group(P<0.05).QiLing medicament could improve the levels of IgM and IgG in Serum,the IgM and IgG level of three QiLing medicament groups were notably different to CY model group(P<0.05),the level of IgA was not of notable difference(P>0.05).The C3 and C4 levels of three QiLing medicament groups were not of notable difference too(P>0.05).(2)IL-2mRNA levels in splenic lymphocyte cells were measured by semi-quantitative RT-PCR method,the result indicated that QiLing medicament could promote IL-2mRNA levels in T cells(P<0.05).The middle dose QiLing medicament was of best effect.(3)Activity of SOD,GSH-Px,T-AOC and the amount of MDA in serum and spleen tissue were detected.The results indicated that:the activity of SOD of spleen tissue was not different to CY model group but the activity of GSn-Px,T-AOC and the amount of MDA of spleen tissue were significantly increased(P<0.05).The activity of GSH-Px,T-AOC in serum of three QiLing medicament groups were significantly increased than CY model group(P<0.05);the amount of MDA in the middle dose group and high dose group were notably different to CY model group (P<0.05).Activity of SOD in serum were not of notable difference(P>0.05).It was concluded that QiLing medicament could improve the antioxidation function of immunodepressed mice.(4)Three QiLing medicament groups could increase the amount of TP,ALB and GLO,and reduce the contents of ALT and AST with significantly different to CY model group(P<0.05). The results showed that QiLing medicament could lighten the damage to liver function by CY.(5)Microstructure and ultramicrostructure of spleen of tested mice were observed which showed that the spleen of CY model group was broken and the white pulp and red pulp structure were destroyed.The white pulp was narrow down to disappear.The lymphocytes were sparse and apoptosis,nuclear chromatin pycnosis and mitochondrial was swelling in spleen.Significant histological changes were observed in mice spleen which showed that QiLing medicament could lighten the damage to spleen function by CY.
