The Study On Tissue Culture And Mutation Of Godetia.grandiflor Lindl

With seed of diploid Godetia.grandiflor lindl,as materials,studied the effect of tissue culture and mutation,To establish the rapid propagation system of Godetia.grandiflor lindl;filtration fine variants,enriched germplasm resources and provided new method for breeding ornamental plants in future.The results were as following:1,Among the five callus induction mediuare(MS,MS+ZT_0.5,MS+TDZ _0.5,MS+2,4-D_0.5,MS+NAA_0.5),explants were inducion callus and differentiation adventitious buds on MS+ZT_0.5and MS+ TDZ_0.5.MS+ZT_0.5were the best mediuare,the rate of diffeentation of adventitious buds was 83%,the best mediure of rooting were 1/2MS+IBA₂.2,Six doses 0Gy,10Gy,50Gy,100Gy,150Gy and 200Gy were used to study the effects of seed irradiation with 60Coγray on the growth of seedling of Godetia grandiflora Lindl.in vitro in this experiment.The results showed that the number of roots and leaves,and plant height all decreased with the increase of irradiating doses(50-200Gy).Moreover,the higher dose,the higher death rate,and the Half Lethal Dosage(LD50)for the seedlings of 41 days was 150 Gy.Mutants were found amongst seedlings in all treatments 70 days after culturing,including flower colour,shape and the structure of patals. This indicats that gamma ray irradiation can increase the frequency of mutations and produces special types of flower in Godetia grandiflora breeding.3,The seed of Godetia.grandiflora Lindl.were impregnated 6h and 24h with difference consistency of colchicine,the rate of death of tissure culture seedling increased with consistency of colchicine increased.Damage caused by lower consistency and longer time were severitier than longer consistency and lower time.the growth potential of tissure culture seedling and shoot difference have the same effect.The experiment results indicated:the doubling plants were inducioned.with 0.4%colchicine impregnat 6h,the rate of doubling were 42%.Leaves of tetraploid seedlings of Godetia.grandiflora Lindl.were larger,thicker and greener.The amongst of flower are colour and shape.Results of the BD FACS Calibur flow cyometric analysis showed that the DNA content of amongst seedlings is chimera of diploid and tetraploid.
