Using the anillary bud of Blumea balsamifera(L.)DC as the explants, the induction and multiplication of bush-bud and adventitious bud,rooting induction and transplanting are systematically studied,to analyze the chemical components of volatile oils from tissue plant and wild plant. Determines the rapid propagation most suitable condition,Discussion use biological technology rapid propagation production medicine feasibility.1.The culture of stem apex inductionThe stem apex of Blumea balsamifera(L.)DC were used as explants and cultivated in the basic medium MS with BA,NAA.MS+BA 2.0 mg/L was most suitable for induction of bush-bud with the 100%rate of induction,The average buds the number was 15.2.Stage of sprout multiplicationThe cluster buds were cut and cultivated in the sub-enrichment culture medium MS adding different concentration of 6-BA,NAA.its best multiplication medium was MS+BA 2.0 mg/L+0.2 mg/L NAA.3.The culture of rootingGrows the vigorous and healthy bud seedling high 3-4 cm to cut individually,cultivated in the rooting medium,the bettermedium for rooting induction is 1/2 MS+NAA 0.5 mg/L.4.Stage of transplantation There were four stages in transplantation,that sand bed prepairing, domestication,pseudoplantation and transplant soil and basin.The survival rate of transplantation can reach 80.87%。5.Extraction of volatile oilTakes the annual leaf separately the tissue plant and the wild plant,the flower uses the methods of HD.Obtains has the special fragrance decadent volatile oil and the crystal mixture,Branches out the crystallization to use in the GC-MS.The tissue plant lower part the adult plant,middle the leaf blade results in the oil rate respectively were 6.68%,4.26%,Lower part the wild adult plant,middle the leaf blade results in the oil rate respectively were 11.29%,9.27%,The tissue plant of flower the oil rate was 1.19%.The experiment proved the volatile oil mainly comes from the leaf,the wild adult plant results in the oil rate to be higher than the tissue plant.6.The Chemical Components of Volatile Oils by GC-MSTo analyze the chemical components of volatile oils from Blumea balsamifera(L.)DC by GC-MS.The tissue plant lower part the plant leaf blade volatile oil GC-MS to separate 8 compound,constituent was:Camphor (68.41%),L-Borneol(17.19%);Lower part the wild plant leaf blade volatile oil GC-MS separates 10 compound,constituent was:Camphor(6.637%), L-Borneol(46.737%);The tissue plant crown flower volatile oil GC-MS to separate 8 compound,constituent was:Camphor(11.13%).The tissue culture technology produces mugwort accepts fragrant mainly includes the camphor, the L-Borneol equicomponent,contains the ingredient with wild not to have the remarkable difference,but has the big difference in the comparative content.Therefore tissue plant whether can replace truly wild to use in also needing the further experiment clinically to carry on the confirmation.
