Some important agronomic traits including plant height, leaf number, fresh weight and yield components were investigated by using a set of 200 RIL population (F7) of Sorghum314×Sudan Grass variance analysis, correlation analysis and stepwise regression analysis; And SSR Markers was used in genetic analysis of 200 RIL population (F7). The main results were showed as follows:1. The tremendous transgressive segregation for all traits was observed in the population and the frequency of these traits was approximately normally distributed. All traits between parents were significantly. The correlations between leaf width, plant height, leaf number, spike length, leaf length and yield were significant, and the regression equation contributed 44.88% of genetic variation of the fresh plant weight. Besides, plant height, leaf number and tiller number separately contributed 14.74%,22.42% and 10.81% of genetic variation of the fresh plant weight.2. A number of factors were studied to set up the optimizing SSR reaction system. The optimized technology was: each 20μL amplification reaction solution comprised 100ng DNA, 4.3mmol/LMgC12, 1.0mmol/L dNTPs, 0.5μmol/L of each Primer,0.75U TaqDNA Polymerase, in addition 10xBuffer 2.5μL. This system had well reproducibility and stability proved by amplification practice.3. The statistical result in the SSR Fingerprint was:①60 segregated lines were identical to female parent, 58 lines were identical to male parent, and 32 heterozygous plants were segregated from the RIL population, and missing were 50 lines;②the purity degree of the RIL population with molecular marker is 59.0%, mixed degree is 16.0%, respectively, and missing was 25%.4. Using plant height as purpose traits, 37 significant differently plants was chosed in the character analysis, 32 heterozygous plants in the SSRs, and 12 plants was identical.The 12 heterozygous plants were studied again with the SSR, 6 heterozygous plants segregated, and rejected and the others homozygous plants was the NIL in plant height.
