| Higher Fungi are abundant sources in isolation of new pharmaceutical products,including the antitumor and metabolition with immunostimulating properties.Many Basidiomycetes fungi contain biologically active compounds both in fruit bodies and cultured mycelia.It is necessary to develop a fermentation process for efficient production of these valuable metabolites. In this study,we isolated mycelium from Suillus flavidus and leucopaxillus giganteus,Their fruit bodies have obvious antitumor effect,and the compounds with bioactivaties have been isolated.The optimum conditions for the bioactive compounds production from cultured mycelia were investigated. The bioactivities of their ethyl acetate,methanol crude extracts from the cultured mycelium were also tested using MTT assay.The results were aummerized as follows:1 the optimum composition of themycelia fermentation medium for S.flavidus is as following:glucose 20.0 g/L,potato 200.0 g/L,yeast extract 7.0 g/L, KH2PO4 3.0 g/L,MgSO4·7H2O 1.5 g/L,VB10.01 mg/L and CaCO3 2.0 g/L.The highest yield of mycelia was obtained under the conditions when the shaker rotation speed 160 r/min,PH 5.8,temperature26℃,and medium loading 600 mL/1000 mL,respectively.2 The ethyl acetate extracts were effective in the suppression of proliferation of human tumor cell lines,including Hela(cervical tumour cell), Bcap37(breast cancer cell),and MCF-7.The bioactive compound was isolated from the mycelium of S.flavidus,which proved to be the suillin.3 The fermentation conditions for leucopaxillus giganteus was set with a basal medium mycelial inoculum containing sucrose 20.0 g/L,,corn5.0 g/L,yeast extract 7.0 g/L,KH2PO4 3.0 g/L,MgSO4·7H2O 1.5 g/L,VB10.01 mg/L and CaCO3 2.0 g/L.The highest yield of mycelia was obtained under the conditions with PH 6, and temperature 25℃. 4 The ethyl acetate extracts of mycelium showed significant cytotoxincity,at IC50=100μg/ml.spectral analysis using HPLC,and ESI-MS revealed the active compound is clitocine or it's isomer. |
