On the basis of the traditional processing drug of Ginseng, this paper aims to explore a new preservation method of fresh Ginseng and established a technology of processing the honey-made red Ginseng by adopting the modern technology of process and combining with the photochemical analysis methods; Based on this, a HPLC fingerprint spectrum of Ginseng extraction will be set up, which paves the way for the establishment of the quality standard for red Ginseng theoretically and methodologically.Therefore, the following were the details about this research and the achievements:1. Comparing to the samples made by vacuum cold storage and the ones made by vacuum freezing, this research figured out the total ginsenoside by colorimetric analysis; tested the solvent of 70%vanallic aldehyde H2SO4 as the colorimeter and the mono-ginsenoside by HPLC. It also established the HPLC grads mode of washing and dehydration with the mobile phase as acetonitrile-water: 0~35min,A:19%;35~55min,A:19~29%;55~70 Min,A:29%;70~100 min,A:29~40; It sets up the regression Rb1:Y=102388.7500X+51.8000 r=0.9996;Re:Y =106718.500X-8324.3000 r=0.9998. The result showed that the best method of fresh Ginseng storage was vacuum freezing.2. According to the standard of"Medicine thoroughly soaked and the honeydew completely absorbed", a technology of processing fresh Ginseng with Chinese traditional medicine juice and honeydew was set up. And after examining the content of the total ginsenoside and the mono-ginsenoside Rb1 and Re, this research found that the best proportion of fresh Ginseng and Chinese traditional medicine juice was 2:1; the best proportion of dry Ginseng and Chinese traditional medicine juice was 1:3.5. Based on these, the red Ginseng Zhengkuo preparation process was already established.3. Based on the HPLC grads mode of washing and dehydration with the mobile phase as acetonitrile-water: 0~35min,A:19%;35~55min,A:19~29%;55~70 Min,A:29%;70~100 min,A:29~40 as well as the examination on the degree of similarities. A HPLC fingerprint spectrum of red Ginseng extraction which concludes eighteen common fingerprint peaks was set up,Several innovations achieved in this research are as following:1. The method of freezing storage was firstly used to preserve fresh Ginseng in order to avoid the affection of preservation fresh chemicals.2. By using Gardenia juice to immersing fresh Ginseng for the first time, a feasible technology of processing honey-made Ginseng was adopted. On this basis, a tasty and healthful product made of red Ginseng Zhengkuo was developed and exploited.3. A HPLC fingerprint spectrum of Red Ginseng extraction is established for the first time.
