The synthesis, characterization and activity test of a new herbicide antidote of dichloroacetyl oxazolidine N-dichloroacetyl-2-methyl-1-oxa-4-aza-spiro-4.4-nonane was described in this paper.一,The synthesis,characterization of N-dichloroacetyl-2-methyl-1-oxa-4-aza-spiro-4.4-nonane1. The materials were isopropanolamine. Cyclopentanone and dichloroacetyl chloride.The compound was synthesized by hydroxide sodium as catalyst. the optimum experimental conditions of the compound by Orthogonal experiment scheme of three factors-three levels were as follows: benzene was solvent,temperature was﹣15~﹣10℃, stirring time was 3 h,the yield was50.0 %,melt temperature was 102~103℃.2. The compound was synthesized by triethylamine as catalyst. the optimum experimental conditions of the compound by Orthogonal experiment scheme of two factors-three levels were as follows: benzene was solvent, temperature was 5~10℃, stirring time was 2 h, the yield was 37.4 %,melt temperature was 101~102℃.3.The structures of compound was confirmed by TLC, UV, IR, 1HNMR,MS and elementary analysis.二,The test of activity of N-dichloroacetyl-2-methyl-1-oxa-4-aza-spiro-4.4-nonaneActivity was test on maize seedlings by planting in water and soil. by testing the change of GSH and ALS in maize seedlings to explain the detoxification mechanism of herbicide antidotes.1. Maize as indicator plant, soaking maize seeds in the compound(0,1,2,4,5,8,10 mg/kg), by raising it in acetochlor water(5,10,20,40,60 mg/kg)and chlorimuron(0.5,1.0,2.0,4.0,6.0μg/kg), test length and fresh weight on foot and new buds .the result of experiment was: soaking maize seeds in the compound, the maize growths was protected differently. when planting maize in acetochlor water ,the concentration of compound 8 mg/kg was the best one. when planting maize in chlorimuron water ,the concentration of compound 5 mg/kg was the best one.2. Maize as indicator plant, soaking maize seeds in the compound(0,1,2,4,5,8,10 mg/kg), by raising it in acetochlor soil(5,10,20,40,60 mg/kg)and chlorimuron(0.5,1.0,2.0,4.0,6.0μg/kg)6 days, test length and fresh weight on foot and seedlings .the result of experiment was: soaking maize seeds in the compound, the maize growths was protected differently. when planting maize in acetochlor soil, the concentration of compound 8 mg/kg was the best one. when planting maize in chlorimuron soil, the concentration of compound 5 mg/kg was the best one.3 Test GSH content and ALS activity of maize seedlings in soil. the result of experiment was: soaking maize seeds in the compound, GSH content and ALS activity of maize seedlings were increased differently, when acetochlor detoxification in soil,GSH was 108.1 % and ALS was 142.9% to comparison under the compound 8 mg/kg;when chlorimuron detoxification in soil,GSH was 106.8 % and ALS was 116.7% to comparison under the compound 5 mg/kg.The result of the experiments indicate: N-dichloroacetyl-2-methyl-1-oxa-4-aza-spiro-4.4-nonane synthesized in this paper can alleviate injury on maize under residues in soil of acetochlor and chlorimuron in different degree.
