In order to investigate the effect of the genes responsible for starch synthesis on cooking quality in rice, near isogenic lines (NILs) were developed , which used an indica variety with poor quality,Guichao2hao, as the recurrent parent and a japonica waxy variety with good quality, Suyunuo, as the donor parent. The gene-tagged molecular markers were used to build the near isogenic lines of starch synthesis related genes both under the non-glutinous background and the glutinous background. Development of near isagenic lines of starch synthesis related genes was started at 2001 in Yangzhou,and till now we already had BC7F2 populations,including NIL-Sssâ… ,NIL-Sssâ…¡-3,NIL-Sssâ…¢-1,NIL-Sssâ…£-1,NIL-Sbe1,NIL-Sbe3,NIL-Isa,NIL-Pul under the non-glutinous background;and NIL-wx-Sssâ… ,NIL-wx-Sssâ…¡-3,NIL-wx-Sssâ…¢-1,NIL-wx-Sbe1,NIL-wx-Sbe3,NIL-wx-Isa under the glutinous background. Starch physicochemical (properties apparent amylose content, gel consistency and the Rapid Visco Analyser profile) of each lines was studied and with the main results as follows:1. In the non-glutinous background:(1) Each candidate individuals has no significant difference with Guichao2hao on most agricultural properties, which indicated that the backgrounds of the NILs and recurrent parent were nearly the same. However, the grain length of most NILs was longer than the recurrent parent. (2) The allelic differences of all amylopectin synthesis related genes between the two rice varieties almost had no significant effect on apparent amylose content (AAC) and the gel consistency.(3) The allelic differences of Sssâ… ,Sssâ…¡-3 and Sssâ…¢-1 had significant effect on most of the RVA profile characteristics. Sssâ…£-1 only made effect on packing temperature. Sbe1,Sbe3,ISA and Pul had little influence on the RVA profile characteristics.2. In the glutinous background:(1) There was no significant difference on the most agriculture properties between the NILs and Guichao2hao, which meant that the backgrounds of the NILs were nearly the same as the recurrent parent. But all the NILs had lower grain weight than the recurrent parent.(2) The allelic differences of all amylopectin synthesis related genes detected between the two rice varieties had no significant effect on the apparent amylose content (AAC).(3) Except Sssâ… the allelic differences of all the other amylopectin synthesis related genes detected between the two rice varieties had no significant effect on the gel consistency.(4) The allelic differences of all the amylopectin synthesis related genes except Sssâ…¢-1 detected between the two rice varieties had significant effect on RVA profile characteristics.
