To Improve wool fineness quality is always one of the important goals for fine sheep breeding. XinJi fine-wool sheep is which can product good quality wool in China, the main types of wool are fine wool, other individuals are the super-fine wool. In order to research the genetic factors causing the difference between fine wool sheep and super fine wool sheep, AFLP techniques were used to analysis genomic DNA, cDNA-AFLP techniques were used to display the mRNAs expression status in skin between two representative sheep group which were the fine wool sheep and super-fine wool sheep.The main results of this study were presented as following:1.The condition of related techniques was optimized by a series of experiment and the silver-staining AFLP experimental system was established. Six pairs AFLP primers were used, genomic DNA from 24 sheeps were restrictived by Taqâ… and EcoRâ… ,226 AFLP markers were obtained, the average obtain marker numbers of individual primer were between 29~50. The results indicated as the following : AFLP marker is suitable for analysis genetic polymorphism in sheep; The similarity index of population in Xin Ji Superfine Wool Sheep was 0.914~0.982.2. In this study ,64 pair primers were used to display the total mRNA of skin Between two wool type sheep, Nearly 1200 cDNAs were examined and 21 differential expression transcript-derived fragments (TDFs) were obtained, of which 16 TDFs were up-regulated in super fine wool sheep group and the others were down-regulated in fine wool sheep group. 8 TDFs were successful cloned and sequenced, By Blast of these sequences in GenBank, Homologous analysis indicated that four of 8 sequences were ignorant ESTs in sheep and the others were acquainted identical to Ovis aries uterine myometrial annexin 2 mRNA, Bos taurus heterogeneous nuclear ribonucleoprotein H2 (H') (HNRPH2) mRNA, Ovis aries high-sulfur keratin BIIIB4 protein (BIIIB4) mRNA, Bos taurus calpastatin (CAST) gene.The study was helpful to understand the molecular differences of wool fineness between super fine wool sheep group and fine wool sheep group. And also increase new materials in sheep genetics and breeding.
