Streptococcus suis type 2 is one of the most important zoonosis, which infect human as well as pigs, horses and cats. It caused permanent deafness or deafness, septicaemia and death. SS2 did cause the big economic loss in swine farming. The pathogenesis of SS2 hadn't been deeply studied so far, and the dynamic distribution and localization of SS2 in vivo hadn't been clear. The study used immunofluorescence to study dynamic distribution and localization of SS2.(1)Establising specific immunofluorescence method to detect SS2 in the tissue. It can detect SS2 directly from the tissues and have feature of specific, sensitive and quickness.(2)Localization of SS2 in SPF pigs's tissues. 30 one-month-old SPF pigs were randomly divided into experiment group (include 20) injected with Sichuan strain ZY05719 and control group (include 10) injected with asepsis medium. The SPF pigs were dissected and inspected for pathological change rules in 12h, 1d, 2d, 3d, 4d, 5d, 6d, 7d, 8d, 10d after injection. Collect the samples of tonsil, lymphoid node, liver, spleen, lung, brain and make frozen sections. The localization of SS2 in vivo organism was detected by immunofluorescence. The study indicated that positive S. suis was found in all identifiable crypt areas; SS2 also appeard in marginal zone and periarterial lymphoid sheath of spleen, hepatic sinusoid of liver, interstitium of lung; presence of SS2 was at the cortex where histopathological lesions and expression of inflammatory mediators were observed.(3)Dynamic distributing of SS2 in SPF pigs's tissues. 30 one-month-old SPF pigs were randomly divided into experiment group (include 20) injected with Sichuan strain ZY05719 and control group (include 10) injected with asepsis medium。The SPF pigs were dissected and inspected for pathological change rules in 12h, 1d, 2d, 3d, 4d, 5d, 6d, 7d, 8d, 10d after injection. The distribution and change rule of SS2 in vivo organism was detected by bacteria isolation and immunofluorescence. The study indicated that there is similar kinetics in the liver, kidney, lung and spleen which the number of bacteria had been rising in 3 days after infection and declined after 6 days; SS2 could be detected in the tonsil at 12h after injection and exist persistently; there is no regular distribution in the brain.
