The present studies show Chlorogenic acid is the most effective component,which has marked activities such asdetoxification,diminish inflammation,antisepsis and so on.The thesis had studied resource about Viola yedoensis Makino,the extraction,isolate and purification process of Chlorogenic acid in the Viola yedoensis Makino by comparative methods,on top of,investigated the biological activity of extractions and identified the composition in the extractions.Results showed:1.Resource about Viola yedoensis MakinoUtilizing the measure of HPLC and UV to identify the composition in the extractions from the Viola yedoensis Makino.From the fingerprint chromatogram we can draw a conclusion that chlorogenic acid is the composition in the extractions.And the method is established for determinating the content Chlorogenic acid in Viola yedoensis Makino.The content of the chlorogenic acid in the four kinds of Violaceae is 19.40mg/g in Viola rossii Hemsl.,17.09mg/g in Viola confuse Champ.,14.09mg/g in Viola philippica Cav.(Viola yedoensis Makino),10.95mg/g in Viola selkirkii Pursh..The content of the chlorogenic acid in the Viola yedoensis Makino is 0.803%in spring,1.082%in summer,0.929%in autumn,0.928%in winter,1.125%in the east of Hunan,0.965%in the south of Hunan,1.082%in the west of Hunan,1.487%in the north of Hunan,1.084%in the middle of Hunan.The highest content of chlorogenic acid in the Viola yedoensis Makino is in summer and in the north of Hunan.2.Extraction process of Chlorogenic acidBy orthogonal design,the optimal conditions of extraction is that the concentration of alcohol is 60%,the ratio of solid to liquid is 1:14,the extracting time of ultrasonic 20 mins,in twice.The content of chlorogenic acid is about 1.487%.3.Isolate & purification process of chlorogenic acidIn terms of the result of static adsorption and static de-adsorption test,H-103 resin is the best one for isolation chlorogenic acid.The optimal adsorption parameters is concentration of feed 0.261mg/mL,pH value 3,feeding rate 3mL/min.The optimal de-adsorption parameters is alcohol concentration 60%,elution rate 3mL/min,elution volume 2BV.The purity of product is 37.69%,the gained rate of chlorogenic acid is 80.72%.
