The oatβ- glucan is a non-starch polysaccharide which is mainly contained in the albumen and aleurone layer cell wall of oat seeds. In this paper, we concerned to theβ- glucan of oat, study on the measuring method, the varying of content, the accumulating rule ofβ-glucan and the formation and development of aleurone layer.Through researching the buffer of the measuring method inβ-glucan, we can learned the pH of succinic acid buffer in the enzyme method is 5.0, of phosphoric acid buffer in the congo red method is 8.0, from these data, we reach the accurate measuring results in experiment. In modified enzyme method, purifying the cellulase is the most important, and if pre-treatment of cellulose is enough, enzyme which is home-products can instead of the import ones. Measurement's error margin is less than 0.1%, however, examining expenses can lower consumedly. By comparing with the centrifuge velocity of dealing with liquids, we educe that the speed of centrifuge is 5000rpm, at 10 min, the result is more ideal. Referring to the research of the varying rule ofβ-glucan and glucose contents in the whole reproduction period of the oat seeds, we inferβ-glucan content is gradually increasing from blossom to seeds matured. In this period, the totalβ-glucan gathers synthesize quickly in 15d after flowering, so it is the key time of theβ-glucan synthesize and accumulation, after this time, the speed of synthesize and accumulation is slowing. The dissolubeβ-glucan gathering velocity is more quickly at 10d after flowering, then accumulating tediously. Theβ-glucan content of the different species oat is quite identical, and the dissolubeβ-glucan comparison perform high in skin oats than in naked oats. From the oat seeds forming to maturity, the glucose content all present a trend that is descending after rising, and at 15d is attain the biggest value, the glucose andβ-glucan content doesn't show the notable relativity, the glucose content isn't the restrict effects ofβ-glucan synthesize.On the trail of research to the motional development of aleurone layer, we initiate to understand the mechanism of aleurone layer formation and the rule ofβ-glucan accumulation, the results manifestate, the oat aleurone layer is 1-2 layer cells, cells gather nourishment material along with grouting carrying on. Theβ-glucan mainly exist in gathers sugar mainly is exist in the aleurone layer and subaleurone layer, it's content is few in seed capsule and inside endosperm.
