Study Of Protective Effect Of Different Dilution Components On Frozen Bull Semen

This experiment was conducted to investigate the quality of Holstein bull which are introduced by Western Inner Mongolia improved cow breeding center,and detected the straw semen quality.The experiment added vitamin B12 besides the existing semen dilution extender,and studied if soybean phosphatide can replace egg yolk in frozen extender for Holstein bull semen.The results showed that:The 12 Holstein Bulls that were introduced by Western Inner Mongolia improved cow breeding center can fit for the requirement of reproductive,and can be used for breeding.The indexes of the straw semen correspond the standard.In adding VB12 2mg/L group ,the activity of the sperm after thawing and sperm acrosomal integrity were higher than control group and the difference were significant(P <0.01); sperm abnormal rate and HYD activity significant lower than the control group (P <0.01); The GOT activity significant lower than the control group (P <0.05). The result showed that: the addition of 2mg/L VB12 group was better.Different dissolution temperature of soybeans phosphatides will influence the protective effect. In adding 2.5%,5.0%,7.5%and 10.0% soybeans phospholipids group, the quality of frozen-thawed sperm is the best when dissolution soybeans phosphatides temperature is 70℃.The concentration of soybeans phospholipids also affects the frozen-thawed sperm quality. The result is good when use 2.5% 5.0% and 7.5% soybeans phospholipid to dilute semen. Adding 5.0% of the phospholipids group was the best of all,in this group the activity of frozen-thawed sperm was significantly higher than yolk dilute group (P <0.01);malformation rate significant lower than other groups (P <0.01). The activity of sperm that dilute by 5.0% soybeans phospholipid within 0h,0.1h and 1h after thawed was significant higher than yolk dilute group (P <0.01).But the difference of sperm activity within 2 and 4h was no significant (P> 0.05). The activity of GOT that dilute by 5.0% and 7.5%phospholipid was significantly lower than yolk dilute group (P <0.05); The activity of hyaluronidase in 5.0% phospholipids group was lower than egg yolk dilute group no significant difference (P> 0.05).Different dilution have different effect to the inorganic elements within seminal plasma . Ca2+, Na+ concentration in phospholipids group was significantly lower than the yolk diluted group (P <0.05). The difference of K+,Cl-concentration between 5.0% phospholipids group and yolk group was no significant (P> 0.05).The difference between other groups and yolk was significant (P <0.05).The in vitro fertilization experiments was carry on using 5.0% soybeans phospholipid diluted straw semen and egg yolk diluted straw semen.The cleavage were 37.66% and 47.18%,blastocysts rate were 14.2% and 18.46%. Blastocyst hatching rate were 31.81% and 38.89%.
