| Soybean is the most important crop for its high protein and oil contents in the world.China is the important origin and home of soybean.The development of molecular markers has brought breakthrough the theoretical and appllied study of soybean.It is widely applied to genome mapping, marker-assisted selection, genetic diversity, genetic distance analysis and varieties identification in soybean and so on. SSR markers are distributed in soybean genome broadly, and it is even, random., more diversified, following Mendelian inheritance.The SSRs are multiallelic codominant genetic marker.At present, the research at pubescence character concerns mainly on cotton, tomato, tea and other crops, but the research on soybean pubescence is not much.Now someone has breeded new glabrous soybean variety with good phenotype as green soy bean. To probe the genetic law of soybean, find the molecular marker linked to it, and quiken the utilization of glabrous character of soybean.Father is glabrous HanShengLuo and mothers are pubescent iLin38, JiLin36, JiKel in this study. The result of hybridize shows that F1 is all glabrous. The segregation rate of glabrous soybean and pubescent soybean of F2 is 3:1. Glabrous character and pubescent character of soybean are controlled by a pair of alle-genes In the homozygotes glabrous character is controlled by a pair of dominant genes and pubescent character is controlled by a pair of recessive genes.Glabrous character is complete dominance to pubescent character,and pubescenct character is recessive character.The optimization SSR reaction system is:(1) 94℃predenaturation 4min, 94℃45s, 55℃annealing for 45s,72℃prolong for 1min,40 circles,72℃prolong 10 min, 4℃forever.(2)The reaction system was amplified in 25ul reaction volumes each containing 10×PCR buffer(25mmol/L) 2.5ul,Taq polyrase(2U/ul) 0.5ul,dNTPS(10mmol/L) 0.4ul, primer(16.8umol/L)1+1ul,genomic DNA(10ng/ul)4ul,sterile pure water 15.6ul.The molecular marker linked to glabrous character is sieved by BSA. By observing on phenotype in field, The glace DNA pool and pubescence DNA pool are composed of the ten individuals respectively. Analyzing the primer showing difference, we find that the SSR marker of Sat020 is linked to the glabrous character. Afterwards, we analyze the primers near the Sat020 belongs to K linkage group Sat126 is found diversified. SSR analysis has been carried out on 122 individuals of F2 generation, and the linkage map is drawn, The genetic distance of Sat126 and Sat020 is 3.6cM and 10.8cM separately. |
