The experiment was conducted to study the effect of oxidative stress on glucosetransport and glucose transporters in piglets'intensine. A total of 12 piglets weredivided into three groups of 4 pigs each group, the first group was control, Se was0.2mg/kg; the second group was low Se oxidative stress, at the beginning of theexperiment, the pigs received an intraperitoneal injection of diquat at 10mg/kg of bodyweight, Se was 0.2mg/kg; the third was high Se oxidative stress, 0.4mg/kg, at thebeginning of the experiment, the pigs received an intraperitoneal injection of diquat at10mg/kg of body weight. the experiment last 49 days, killed all pigs, separatedintensine, scraped intensine mucosa. Activities of SOD,XOD,GSH-PX and content ofMDA in intensine mucosa tested were to identify oxidative stress. BBMV and BLMV wereprepared by an MgCl2 precipitation, BBMV and BLMV marker enzymes determinatedwere to identify the qualities, The uptake of D-glucose was measured at 37℃by a rapidfiltration technique for time periods ranging from 1 s to 30min; Reverse transcriptionPolymerase Chain Reaction(RT-PCR) was applied to determinated the expressions ofNa+-glucose transporter 1 mRNA and glucose transporter 2 mRNA. The result was asfollowed:Group2 or group 3 compared to groupl, activitives of SOD and GSH-PX decreased(P<0.01), MDA content and XOD activity increased (P<0.01), group 3 compared togroup 2, XOD activity increased (P<0.05);The transport of glucose in each group increased with increasing of Na+ level, according to the regression curse, the transport of glucose in groupl is maximum(147.70)at the 138.3 mM of Na+ concentration, the transport of glucose in group2 is maximum(109.52) at the 259.83 mM of Na+ concentration; the transport of glucose in group 3 ismaximum (104.09) at the 181.70 mM of Na+ concentration; Compared to groupl, at the 0.1 mM of glucose concentration, the transport of glucosein BBMV or BLMV decrease(P<0.01), at the 1 mM of glucose concentration, thetransport of glucose in BBMV or BLMV decrease(P<0.01); group 3 compared to group2, at the 1 mM of glucose concentration, the transport of glucose in BBMV or BLMVincrease (P<0.05); Group2 or group 3 compared to groupl, the expressions ofNa+-glucose transporter 1 mRNA and glucose transporter 2 mRNA decreased(P<0.01).Conclusion:①Oxidative stress impaired intensinal mucosa and supressd the activities of themark enzymes;②Oxidative stress inhibits the transport of glucose in BBMV or BLMV of intensinemucosa, it associated with decreased the expressions of SGLTlmRNA or GLUT2 mRNA;③Under oxidative stress, high concentration of Na+ can improve the transport ofglucose in BBMV of piglets'intensine mucosa;④0.4mg/kg Se may resist the oxidative stress of an intraperitoneal injection of diquatat 10mg/kg of body weight, but the function isn't obvious.
