Studies On The Preparation And Pharmacologic Action Of Solanum Lyratum Thunb Polysaccharides Injection

【Background】Under the modern cultivation condition, the occurrence and prevalence of the animal'sbacterium and the virulent diseases presents some new characteristics, and also brings newdifficulties for the animal epidemic disease's prevention and cure, although the westernmedicine's prevention and cure has obtained a certain effect, the medicine abuse has also ledto the drug resistance, drug residue, environmental pollution and so on, so seeking anddeveloping the new medicine in the present stage has become the key point of thestockbreeding research.The Chinese medicinal herbs becomes the direction which the multitudinous scholarstudies, because of its lower poisonous and side effect, lower drug residue and difficultlymaking bacteria tolerant to the drug. Our country has the rich resources of Chinese medicinalherbs, but has not always obtain rational development and the full use, specially insufficientlyis also through to the Chinese medicinal herbs' effective components and the Pharmacologyresearch.Therefore, reasonably developing the Chinese medicinal herbs' preparation has thevital significance for the prevention and cure of animal diseases.But there was few report toprepare and study on antibacterial effect and antiviral effect about the Solanum Lyratum ThunbPolysaccharides injection. There was no report to study the mechanism of enhancing immunityabout Solanum Lyratum Thunb Polysaccharides injection as immunoenhancer.【Objects】(1)To prepare Solanum Lyratum Thunb Polysaccharides injection and to measure safety bythe standard of toxic evaluation.(2)To measure antibacterial effect and antiviral effect of Solanum Lyratum ThunbPolysaccharides injection.(3)To study on immune function effection of Solanum Lyratum Thunb Polysacharideinjection for chicks.【Methods】(1)The polysaccharides is extracted from Solanum Lyratum Thunb by way ofhydroalcoholic, Solanum Lyratum Thunb Polysaccharides injection was prepared by way ofpreparation regulations and measured safety by the standard of toxic evalution. (2)Antibacterial effect of Solanum Lyratum Thunb polysaccharides injection was studiedby cup-plate method,minimal inhibitory concentration (MIC) and minimal bactericidalconcentration(MBC) of Solanum Lyratum Thunb polysaccharides injection was measured bydouble dilution tests. Antiviral effect was measured by tissuecell culture.(3)Sanhuang Chicks were divided into five groups: experimental groups(ⅠⅡⅢ)wereinjected Solanum Lyratum Thunb polysaccharides injection andⅤgroup were injectedphysiologic saline.The five groups were inoculated NDV inactive vaccine for 0.2ml/1 only. Atthe different time, samples were prepared for histological section and the index of immunitywere examined by a series of experimental method.【Results】(1) Solanum Lyratum Thunb polysaccharides injection was proved to be a brown-yellowpellucid fluid whose PH value was consistented with requirement.The result suggested that there wereno toxicity,pyrogen and stimulation in the preparation.And a further study showed thatintramuscular injection was safe and reliable.(2)The results shows that Solanum Lyratum Thunb polysaccharides injection have goodantibacterial effect against E.coli,streptococcus, Staphylococcus aureus and Salmonella.TheMIC of Solanum Lyratum Thunb polysaccharides injection is 5mg/ml, 2.5mg/ml, 1.25mg/ml,2.5mg/ml. The results shows that Solanum Lyratum Thunb polysaccharides injection havebetter antiviral effect.(3)The results shows that Solanum Lyratum Thunb polysaccharides injection can enhanceimmune function in chicks.【Conclusions】The prepared method of Solanum Lyratum Thunb polysaccharides injection is simple andstable. The preparation is safe and reliable with the way of intramuscular injection.Pharmacodynamics test indicats: the preparation has remarkable antibacterial and antiviral effect,itcan enhance immunity function in chicks and be used as immunoenhancer.
