The 350 1-old-day AA broilers were randomly divided into a control group and 4 treatment groups of A,B,C and D.Groups A,B,C and D were supplied with 10,20,30 and 40 g/kg of the bitter bean seed's dregs,respectively.On 7,14,21,28,35,42-old-day, 10 AA broilers per group were killed and 0.5g of cecal contents were collected to isolate, incubate and enumerate bifidobacteria by selective media and plate counting methods.Then studies were made of biological characteristics of isolated Bifidobacterium strains by using microscope, scanning electron microscope and gas chromatographic technique,of nucleotide sequence identity rates of the 16S rDNA gene from isolated Bifidobacterium strains via molecular approaches,of the utilization on X-Gal in isolation and enumeration of bifidobacteria.The results showed that the bifidobacteria quantity in the cacal contents of AA broilers was related to the concentration of Bitter Bean Seed's Dregs,and the supplement with 20g/kg of bitter bean seed's dregs to basic diet can increase bifidobacteria quantity in the cecal contents of AA broilers.17 strains were identified through the physiological and biochemical characteristics,and there were 6 B.breve, 4 B.animalis, 4 B.gallinarum,2 B.pseudolongum and 1 B.adolescentis.The ratio of acetic acid to lactic acid exceeded 1 generated from isolated Bifidobacterium strains B2, B4, B6, B9, B15 and B16,and lactic acid totally was L-lactic acid.Nucleotide sequence identity rates of 16S rDNA gene was 97.2% between strain B2 and Bifidobacterium gallinarum, 95.9% between strain B16 and Bifidobacterium animalis,Bifidobacterium animalis subsp.lactis.Furthermore, the relation was closer between strain B2 and B.gallinarum,and strain B16 was intimate with B.animalis and B.animalis subsp.lactis.X-Gal has no identification among Bifidobacterium species in the cecal contents of AA broilers.
