The research of Phellodendron Chinense Schneid, specially the research of the chemical composition was started in 1926, which was carrying on until today, and its quality specification system constantly consummate. At present, the commodity raw material for medicine is to be the object for the research of Phellodendron Chinense Schneid, of which the origin is complex and can't fully reflection trueborn medicine. On the other hand ,the majority research was carried on for the Chemical composition. In the consulting, more than 900 literatures, the chemical Composition research report is more than 600.This research, which was based on truebom medicine, the standard sampling operation, and the statistical analysis method, taking the berberine content as the determination index. In this research for Phellodendron Chinense Schneid, the different recovery years and months, and the different processing method has been carried on for the optimized choice; The feasibility of the low recovery years and the high density plant was researched; Through the relationship research of the outward appearance characters and the intrinsic quality, the outward appearance characters that were used by tradition and folk to appraise the intrinsic quality of Phellodendron .Chinense Schneid was examined its accuracy; Meanwhile, the influence of the skinned regeneration to the content of berberine was preliminarily studied. The results as follow:(1)The Berberine hydrochloric hydrochloric content of Phellodendron Chinense Schneid the years of which are 3--25 years (3, 4, 5, 8, 10, 11, 12, 15, 17, 20yr) have been analyzed in this paper. It showed that the Berberine hydrochloric content was between 58.7—82.5mg.g-1 and the 3yr trees was the lowest and 10 was the highest which the Berberine hydrochloric content were 58.7 mg.g-1 and 82.5 mg.g-1 respectively. The Berberine hydrochloric content was increased according to the tree's age from 3—10 years and it reached the highest in 10 year and begun to decrease after 10 years, and it is consistent with the traditional method.Although the Berberine hydrochloric content reached the highest value in 10 years according to the SSR test, the Berberine hydrochloric content between 8—13 tree age was insignificant, and the variation value was only 1.8mg.g-1. So in order to satisfy the market necessary and shorten the planting time to enhance the economic benefit, the harvest year was brought forward and fixed in 8 to 10years.(2) The water-content ratio of Phellodendron Chinese Schneid which were 8, 10, 12 years old were high in May, Jun and July, it were 59.01%,58.42% and 61.09%, 54.95%,58.82% and 55.6%, 57.05%,53.48% and 58.96% respectively.(3) The Berberine hydrochloric content of 8, 10, 12years old trees was highest in Jun, July and Aug, which the content were 81.4mg.g-1, 82.5mg.g-1, 82.8 mg.g-1 respectively. The Berberine hydrochloric content of 8, 10, 12 years old trees after July had the trend of decreasing. According to the statistics analysis and the SSR test, there was a damned significant relationship between difference harvest month and Berberine hydrochloric content and the Berberine hydrochloric content of June and July was difference significantly with May, Aug, Sep, Oct and November. The highest Berberine hydrochloric content of 8, 10 12 years old were frequently in Jun or July, so the best harvest month of 8, 10, 12 years old trees were fixed in Jun or July.(4) During May to Nov, the Berberine hydrochloric content of female in Jun, July and April of 8, 10, 12 years old trees was half of that in May, and in September it restored to the level of May. But the difference of Berberine hydrochloric content of male trees were not significant and the Berberine hydrochloric content was tranquilization. It was significantly difference between male trees and female trees and the year-average level of Berberine hydrochloric content of 8 years old male and female trees was 20.53mg.g-1, and 10. 12 years old trees were 18.2 mg.g-1 and 17.52 mg.g-1 respectively.(5) The Berberine hydrochloric content of 3 years old Phellodendron Chinense Schneid in main planting district(Hong'ya, Ying'jing, Da'yi, Guang'yuan, Da'zhou, Mu'chuan) in Sichuan province was between 40.2 mg.g-1 and 58.7 mg.g-1, and it was the highest in Ying'jing which the Berberine hydrochloric content was 58.7 mg.g-1. The next district were Mu'chuan, Hong'ya, Da'yi and Da'zhou which the Berberine hydrochloric content were 56.4, 50.2, 48.8, 47.6 mg.g-1 respectively. Because of the highest Berberine hydrochloric content in Ying'jing and there was significant difference in 0.05 level of Berberine hydrochloric conten between Ying'jing and other planting district in Sichuan, so the bark of Phellodendron Chinense Schneid in Ying'jing was trueborn medicinal materials. The bark of 3 years old Phellodendron Chinense Schneid completely conformed the standard of《People's Republic of China Codex》in 2005 (content>30mg.g-1),At Ying'jing trial field, the. average tree circumference and Berberine hydrochloric content of 5 years old Phellodendron Chinense Schneid in three planting densities (3000trees/hm2, 4500trees/hm2 and 6000/hm2) were decreased as well as the increase of planting density and the average trees circumference were 23.05, 19.31, 18.43em and the average Berberine hydrochloric content were 66.2, 63.9 and 62.1 mg.g-1 There was significant relationship between planting density and it's diameter and average Berberine hydrochloric content. According to the SSR analysis, there was significantly difference in 0.01 level of trees circumference and the average Berberine hydrochloric content in the different three planting densities.The bark of 3 years old Phellodendron Chinense Schneid completely conformed the standard of《People's Republic of China Codex》in 2005 (content>30mg.g-1). Alth0ugh it was significant difference between 6000 trees/hm2 and 3000 trees/hm2 of 5 years old Phellodendron Chinense Schneid, the difference value was only 4.1mg.g-1. In order to solve the problem of provision deficiency and satisfy the necessary of Berberine hydrochloric in pharmacy and industry and shorten the planting time to get more economic benefit, it was feasible to adopt the harvest and planting method of less-year harvest and high density planting.(6) The six index (bark thickness, tree's height, chest circumference, roughness degree of bark, leaf type, inner epidermis color) in this experiment had different influence on Berberine hydrochloric content of Phellodendron Chinense Schneid and the relation coefficient arranged from big to small as followings: inner epidermis color(0.77)>bark thickness(0.73)>chest circumference(0.279)>tree height(0.13)>roughnes degree of bark (0.12)>leaf type (0.046). According to the coefficient analysis, the three factors which influenced the qualities were inner epidermis color, bark thickness, chest diameter and they were arranged from big to small: inner epidermis color(0.55)>bark thickness(0.29)>chest circumference(0.09).The inner epidermis color, bark thickness and chest diameter were the three index which the inflection of other outward appearance characters and the three index were the main appearance character index to judge the quality of Phellodendron Chinense Schneid and must given more concentration on it. The darker of inner epidermis color, the thicker of bark and the bigger of chest diameter, the better of the quality, this was correlated to the traditional evaluation index of "thicker bark, darker color were better"(7) According to the survey of the original and regeneration bark, there was the biggest variation value of 5.3mg.g-1between 5 years old original bark and 2 years old regeneration bark and the next was 1.3mg.g-1 which was between 11 years old original bark and 3 years old regeneration bark. The least variation value was 0.6mg.g-1 which was between 13 original bark and regeneration bark. According to the variance analysis, exception of the significant difference between 5 years old original bark and 2 years old regeneration bark, there were no significant difference between 11 and 13 years old bark. The regeneration bark could be the raw material for the extraction of Berberine hydrochloric and this had important practical significance for the sustainable utilization of Phellodendron Chinense Schneid.There were significant correlations between trees age and original bark and the Berberine hydrochloric content of regeneration bark, but there was no significant correlation with bark type (original and regeneration bark), so on the disposal of decortication to the Phellodendron Chinense Schneid, the older trees were the best choice. (9) According to the screening of different extraction reagent and time, 1% hydrochloric, distilled water and anhydrous ethanol were utilizated, the best extraction reagent, for the extraction of fresh and dry sample of Phellodendron ChinenseSchneid and the Berberine hydrochloric content was 77.2mg.g-1 and 70.2 mg.g-1 respectively. And the optimal extraction time was 90rain using two methods. The Berberin hydrochloric content which extracted using l%hydrochloric as the reagent for fresh, sample was higher of 7.2mg.g-1 than that of using anhydrous ethanol as the reagent. Utilization of 1% hydrochloric as the extraction reagent could reach the most extraction, ratio and achieve the efficient utility ofPhellodendron Chinense Sehneid resource.Among the temperature of 40℃—100℃, the Berberine hydrochloric content was decreased as well as the increasing of the dying temperature and the content was 63.2mg.g-1 and 45.2 mg.g-1 and the variance was 18mg.g-1 The best temperature of drying Phellodendron Chinense Sehneid was 40℃.
