Gladiolus"Ovutie","Advance red","Spic and span"was irradiated by 60Co—γwith 0Gy,25Gy,50Gy,75Gy,100Gy,150Gy. Its main quality indices and DNA were observed.The results showed: Its main quality indices were influenced.The results showed that the variation range of main quality indices of plants from irradiated was proportional to the dose of gamma rays. In other words, the higher the dose of gamma ray used,the wider the variation range of the main quality indices of plants was. The corms was beforehand germinated under the dose of 25-75GY. The variation of corm's sprouting rate was not obviously. It was better for the plant growth at 25GY,the irradiation at the does of 75GY gain the most variation, and at 100-150GY the growth indices all goes down obviously.Through RAPD analysis of the Gladiolus radiated with different 60Co—γray dosages ,the results indicated that the great variation of DNA sequence of mutagenic agent might be existent in Gladiolus genome. It implied that RAPD technique provided a new alternative for checking irradiation-induced effects. The optimum concentration of RAPD's five important components was:primer (10μg/L)20ng,dNTPs(2.5mmol/L)2.0μL, MgCl2 (25mmol/L) 2.2μL, 10*Reaction Buffer 2.5μL,Taq 0.3μL, DNA (20ng/μL) 20ng,ddH2O15μL,respectively.Modified thermal profile consisted of all initial denaturation step at 94℃for 10 min, followed by 40cycles of 94℃for 30s, 37℃for 60s. and 72℃for 90s and a final exposure to 72℃f0r 5 min.And then 4℃storage.Of the 12 random primers analyzed in this research, 9 primers can show us steady and polymorphic amplification profiles. The genotype similarity coefficient of the"Ovutie"varieties between 50%-92.3%. The genotype simple matching coefficients ( Ssm ) of the"Ovutie"varieties between0.333-0.464; The genotype similarity coefficient of the"Advance red"varieties between 60%-93.3%. The genotype simple matching coefficients ( Ssm ) of the"Advance red"varieties between0.357-0.469; The genotype similarity coefficient of the"Spic and span"varieties between 44.5 %- 94.1 % . The genotype simple matching coefficients ( Ssm ) of the"Spic and span"varieties between0.344-0.472.The"Spic and span"is the most sensitive of radiation.
