We did the research on cDNA clone of blue color formation-related genes from petals of different development phases in Petunia hybrida,Hyacinthus orientalis and Muscari botryoides and construction of expression vector. Several achievements obtained were as follows:1. We established the improved CTAB method available for extraction of total RNA from saccharide-riched tissues of Muscari botryoides. We isolated Flavonoid-3',5'-hydroxylase genes HF1(1622bp) and HF2(1677bp) which controlled formation of flower color from blue petals of Petunia hybrida, also B5 gene DifF(464bp) which enhanced the activity of F3'5'H.2. We cloned cDNA fragment of 710 bp from Hyacinthus orientalis, this fragment had high homology with F3'5'H in other species through blastx analysis respectively, the homology was 40%-68%. It shown that we cloned cDNA fragment of F3'5'H (accession number: EF468472).3. We constructed plant expression vector (pWR306-HF1) harboring F3'5'H of Petunia hybrida which was available for transformation of Lilium.
