A MADS box gene HoSEPlgrouped to Arabidopsis SEP clade was isolated from regenerated floral bud material of Hyacinthus based on PCR strategy. Analysis of the protein sequence suggest that HoSEPl is most similar to Arabidopsis AGL6 with 64% identity, and by highest identity to two unknown genes, pAOM3 of Asparagus and an OsMADS of Oryza (respectively 90%, 70%) among monocotyledon plants.To characterize HoSEPlgene ,we constructed binary vector , transtormed it into Petunia and Arabidopsis, comfirmed transgenic plants with RT-PCR and analyzed phenotypes of transgenic plants cultured in vivo. Under the long-day conditions the transgenic plants flower earlier than their wild types. From the phenotypes, the transgenic Petunia plants cultured in soil medium show flowering more than 30 days earlier than wild type. The transgenic Arabidopsis plants also show extreme earlier flowering: after sowed 14 d and producing 3 leaves, the plant shorter than 1 cm terminated at a flower. Single HoSEPl transgenic plants converted their leaves to petal-like in vitro, and the leaves of weak phenotypes produce abnormal calli, and further bud differentiation more difficult, such a transformation also confirmed by RT-PCR detection of FBP1 expression, a Petunia gene specific expression to corolla and anthers Petal-like structures, rod-like structures (probable anthers ), ovary-like structures and buds can produced directly from the transgenic calli on bud differentiation medium (BDM). These ovary-like structures also can be produced from the shoot terminal of transgenic plants and calli on rooting medium (RM). In addition, of transgenic plants reduces Fecundity severely and calyx show abnormalily, such as great different from sizes and so on.
