Genetic diversity analysis of different populations of common carp of Ganzhouwas analyzed by random amplified polymorphic DNA (RAPD) assay. The commoncarp were from Rijin county population (RJ), Xingguo county population (XG),Ganxian county population (GX), Yudu county population (YD) and Huichangcounty population (HC). 11 primers that can get the clear and polymorphic bandswere selected in 25 primers. According to the Similar Identity (SI) and SimilarDistance (SD), the relationship of the five populations in Ganzhou region wasmade. 11 primers can get 1772 clear and polymorphic bands. The average percentageof polymorphic loci to total DNA of The five populations was 85.04%. A relativelyhigh level of intraspecific genetic diversity was revealed: He=0.27088, I=0.39562and PPB=66.20% at population level; He=0.3232, I=0.4789 and PPB=85.01% atspecies level. This study demonstrated that the level of genetic variation of the RiJinpopulation (RJ) (H=0.2919; I=0.4241; PPB=70.3%) was higher than that of the otherpopulations.Genetic distance UPGMA system occurrence tree clustering result display,Genetic distance (SD) ranged from 0.1371 to 0.2192, with average of 0.17636, andGenetic identity (SI) ranged from 0.8032 to 0.8719, with average of 0.8418. Nei'sgenetic diversity analysis showed a low frequency of gene flow (Gst=0.3217;Nm=1.0541) existed. Analysis indicated that there was a certain geneticdifferentiation between these populations. The relationship between RiJin countypopulation(RJ) and Xingguo county population(XG) is closest. The relationshipbetween Rijin county population(RJ) and Huichang county population(HC) is farthest.The experimental result shows that, the geographical isolation is the main influence togenetic differentiation. |
