Chinese Jujube and wild jujube are important fruit of our country, but the cultivation of jujube varieties is restricted severely by the traditional cultivation and propagation. It needs to step up the micropropagation study of Chinese Jujube and wild jujube to accelerate the speed of varieties cultivation and promotion.Junzao (including'Junzao','Xingguang','Jiao 2'and'Jiao 5'), Pozao (including'Tangxing','Fuxing'and'Pozao'),'Fuxiang','Dongzao', wild Jujube ('Xing 6') and wild Jujube ('No. 4') diploid, tetraploid, and octoploid used as material, the general conditions of jujube of different genotypes initial culture was studied;the effect of plant growth regulators and concentration on the proliferation of Jujube and wild jujube; using orthogonal experimental design, the effect of different high concentrations of IBA and treatment days on'Dongzao'and wild Jujube (No. 4) diploid, tetraploid, and octopliod was studied with a two-step method. The main results are as follows:1. Initial cultureThe early growth stems of the Jujube and Wild Jujube in May was most appropriate to initial culture, then sterilized with 0.1% HgCl2 for 4-6min and cultured in MS + sucrose 30g/L + agar 3.5g/L + carbon 1g/L, which can reduce the browning rate to be less than 20%, and the survival rate can be higher than 80%.2. Subculture(1) The factors of Junzao proliferation were 6-BA> IBA> strains. The best combination was 6-BA 0.5 mg/L + IBA 0.1mg/L, the propagation index was 2.55. And there was no significant difference among strains, so different strains of Junzao can use the same kind of subculture medium: MS + sucrose 30g/L + agar 3.5g/L + BA 0.5mg/L + IBA 0.1mg/L.(2) The best subculture medium for'Dongzao'was 3/2MS + sugar 30g/L + agar 3.5g/L + 6-BA 0.5 2.0mg/L + IBA 0.1 0.4mg/L, and the propagation index was better than 3.30. Subculture medium for Wild Jujube (Xing 6) was: 1/2MS + sugar 30g/L + agar 3.5g/L + 6-BA 0.5 1.0mg/L + IBA 0.1 0.4mg/L. The propagation index was better than 1.82.(3) Compared the optimal medium for proliferation of Wild Jujube tetraploid (6-BA 1.0mg/L + GA3 0.1mg/L) and diploid (6-BA 1.5mg/L + GA3 0.3mg/L), the used of 6-BA and GA3 concentrations both were reduced; octoploid training required the lower concentration of 6-BA and added GA3 was ineffective, the optimal subculture medium was 6-BA 0.2mg/L + IBA 0.1mg/L.3. Rooting(1) The effect of IBA concentration on rooting of'Dongzao'be slightly larger than the length of treatment time. The best conbination for the rooting of'Dongzao'was pretreatment for 2d with 1/2MS + sugar 30g/L + agar 3.5g/L + 10mg/L IBA, then be transferred to the medium without plant growth regulators to root, the rooting rate can reach 90.56%.(2) The concentration level of IBA was the crucial factor of rooting for wild jujube diploid, but the length of pretreatment time played a bigger work for tetraploid and octopliod. The best treatment combination for rooting of diploid and tetraploid is that pretreating with 15mg/L IBA for 1d and 2d, then transfered to root medium with IBA 0.5 mg/L, and the rooting rate can achieve 93.33% and 96.67% separately. The rooting rate of Wild Jujube Octoploid can achieve 56.67 % by pretreatment with 15mg/L IBA for 1d followed by transfer to root medium with IBA 0.5 mg/L.(3) Rooted in the best pretreatment conditions of polyploidy, we did single factor test. The best concentration of sucrose for Wild Jujube polyploid rooting was 20g/L. Wild Jujube tetraploid was well for direct rooting without dark culture, Octoploid could increase the rooting rate to 78.33% which was cultured under dark for 5d.
