Aprostocetus prolixus has inhibitory action to Apriona germari, and itsparasitism rate may amount to 50ï¼…highly in the open air. Therefore, protection andbreeding Aprostoeetus prolixus have higher application value. But how to strengthenits efficacy of biology prevention is an exigent problem at present. We use PCRmolecular detection technology, RAPD-PCR technology, discontinuous SDSperpendicularity board polypropylene gel electrophoresis and so on, to study therelationship between Wolbachia and Aprostocetus prolixus, the relationship betweenAprostocetus prolixus and Apriona germari, the relationship between Aprostocetusprolixus and other populations. The data were analysed and we got some conclusions.1.Based on the amplification of part of the wsp, ftsZ and 16S rDNA gene, weexpanded the fragment of the 600 bp, 1000 bp and 900 bp respectively in the DNA ofa laboratory population of Corcyra cephalonica (Stainton),which proved theCorcyra cephalonica (Stainton) infected with the Wolbachia; But we did not expandany fragment in the DNA of the Aprostocetus prolixus. So we can infer that theAprostocetusprolixus was not infected by Wolbachia or the infection rate was low.2.The genetic relationship of 6 Aprostocetus prolixus populations from 3provinces (Hebei, Shandong, Hubei) was detected with RAPD markers. Adendrogram was constructed based on RAPD data, the 6 populations are clustered 2kinds. The first kind contains 5 populations, may divide into 3 sub kinds, and: 1 subkind includes Baoding population and Tangxian population, 2 sub kind have Qian Anpopulation only, 3 sub kind include Yixian population and Shandong population (TaiAn), 1 sub kind and 2 sub kind from Hebei Province among them, equally; Yixian in3 sub kind originate from Hebei Province, Tai An originates from Shandong Province.The second kind contains only one population, is Wuchang population, originatesfrom Hubei province.3.From the photograph of esterase electrophoresis, the six geographicalpopulations of Aprostocetus prolixus respectively shows 9,10,8,8,8 and 7 belts.And the most belts is 10,is Tangxian population; the fewest belts is 7,is Hubeipopulation (Wuchang).In these belts, there are 6 location belts are common, but 1location belt of Tangxian population is wider, and Hubei population is deeper. Having4 location belts are proper population belt, 1 location belt is Tangxian population, thecolour is deeper; another 3 is Hubei population, one colour is deeper, the other 2 isabsent. The result indicates that the Hubei population is different from otherpopulations.4.Protein in the living things has a big change, having reported the whole process to a certain extent. We use SDS polypropylene gel electrophoresis method to analysethe protein change in the egg of Apriona germari when Aprostocetus prolixusparasitism over it. From the photograph of protein, Aprostocetus prolixus parasitismmakes the egg's protein change, even change more complex. The first 3 days, theprotein belts begin to change comparatively obvious, just parasitic eggs appear 3 belts,and not parasitic eggs don't appear any belts. The 3 belts disappear right away inthe second day. This period, the protein about parasitism and resists parasitismappearing chance is big.
