Comparison study was done on leaves structure and protein of Stylosanthes accessions resistant and susceptible anthracnose. Two accessions of Stylosanthes Mineirao and CIAT2312 were used to study in this paper.The reaction of detached leaves Stylosanthes of to Collectotrichum gloeosporioides was tested. The stylo leaves of Mineirao were hypersusceptible in seven days, while stylo leaves of CIAT2312 were rotted in the same time. It is speculated that Mineirao may contain substance of inhibit the growth of C. gloeosporioides. The soluble protein of Stylosanthes accessions resistant and susceptible anthracnose was no function to C. gloeosporioides.Leaves structure of two accessions was observed with method of paraffin section. The epidermis cells of Mineirao are thicker than that of CIAT2312. And stoma density of Mineirao is fewer than that of CIAT2312. This result showed that the Stylo resistance may relate to epidermis cells thickness and stoma density.SDS-PAGE was used to study the soluble protein and whole protein of leaves from Mineirao and CIAT2312. The number and band size of soluble protein from Mineirao are same as CIAT2312. Two-dimensional electrophonesis was used to analyse the different compositions between resistant and susceptible accessions, There is one different spot between two accessions from Coomassie Brilliant Blue-stained gel, and three spots between two accessions from silver-stained gel. These different spots may relate to the anthracnose resistant.The infection of Stylosanthes anthracnose resulted in Phenylalanine Ammonial- yase(PAL) and Polyphenoloxidase(PPO) and Superoxide Dismutase(SOD), activities of these three enzymes were increased in Mineirao firstly, while in CIAT2312, they were not similar as that in Mineirao.Chlorophyll content was analysed young plant between two accessions, CIAT2312 is higher than that of Mineirao.
