| Cut flowers of lilium.spp are popular in the flower market in recent year, which has had the second quantity of sales in the flowers' auction market in Holland.In china, lilium cut flowers have widespread availability in many celebrations such as matrimony because of its beautiful implied meanings and its graceful posture. In consideration of lily's commercial potential and the press for its technique of refreshing, this experiment has studied the new fresh-keeping agent's effect on the cut flowers of lilium and the method of 1-MCP'S application; In the other hand, the protein pattern in the petal of lilium were analyzed by SDS-PAGE technique to express the process of lilium's post harvest senescence in protein level. In addition, ultra structure of lilium petal was used to explain the cut flower's senescence. The main results in this experiment were as follows:1.1-MCPand ETP are treated mixed as fresh-keeping agents in this experiment, adopting different concentration and different time between mechanism of flower development of lilium. It is in order to study and analyze the 1-MCP'S effect on refreshing and blocking ethylene synthesis totally. Studies showed 1-MCP (1.2mg/L) had significant effect on all the five varieties lily's refreshing .It also greatly raised the ornamental value of five varieties lilium.spp cut flowers; Treatment of 1-MCP raised the activities of SOD and CAT, and bring down the content of MDA, reduced the phenomenon of lily petal's falling out. It also revealed that 1-MCP treatment was effective in any development of lily's mechanism and it can be used continually during lily's postharvast. 1-MCP treatment could competitive binding to the ethylene receptor and block ethylene synthesis.2.This experiment has determined the physiological index correlated with senescence to studied the relations between the protective enzyme system and fat layer's over-oxidation during lilium's process of senescence which could provide some information for selection of lilium's breeding and refreshing.3.This experiment studied and analyzed the protein change of petal's senescence by SDS-PAGE technology. It established the SDS-PAGE experimental system suitable for lilium.spp protein. It has analyzed the result of SDS-PAGE by computer-assisted image analysis software .It showed the five different varieties lilium.spp have some similar protein bands but it also had clear diversity between different varieties. During the process of senescence, some protein decreased, some held the line. In the declination of petal, new band had appealed in the period of wilting which may be a protein correlated with senescence.4.The process of flower senescence is a ordered death process which was influenced by genetic and intrinsic or extrinsic factors, attending by degradation of some macromolecular substance and the morphological change of petal's cells. The results from ultra-structure to bud,blooming,wilting of Lilium 'Oriental Gr. Sorbonne' petal showed that the organelles of petal cell were losing in order. |
