Xinjiang, an important base of sheep, owing the most, is the important product-place of fine-wool and in the leading place in China. But now, it is difficult to raise or maintain the quality of present superior fine-wool, since it is studied less about the quantitative trait loci (QTL), controlling the main economy traits, which caused hardly selecting the superior fine-wool breed, and furthermore, the decline of quality of wool traits are occurred easily. In order to overcome these problems, it is necessary to study the linkage analysis or QTL study for wool traits of the Chinese Merino (Xinjiang army westland reclamation model) in Xinjiang. According to the above, regard the wool fibre diameter as main character, this paper constructed the reference family of chinese merino sheep by pedigree, and made linkage analysis of wool traits and microsatellite loci.This experiment tested wool information of four economy traits as fibre stretched length, fiber diameter, and birth weight of lamb of tested sheep.According to GENBANK, we selected nine microsatellite loci on ovine chromosome 1th and one on chromosome 3th, amplified the genomic DNA, show amplification results by polyacrylamide gel electrophoresis and silver staining. After determined genotypes, analyse the genetic polymorphism, handled phenotypic dates by analysis of genotypic variance by SPSS13.0, after different genotypes analysis and multipple comparison, draw conclusions as follow:(1)Excepts BMS772(PIC =0.2567)and BM6506(PIC =0.4697)are mid- polymorphic, the rest of microsatellite loci are high polymorphic. The average of population PIC,H,E,Rh were 0.5848,0.6333,3.0315 and 0.4841 respectively, which shows this population have high genetic diversity and medium rate of homozygote of alleles.(2)Microsatellite marker BM6506 and OARDB6 linkage closely with the wool diameter, BM6506 and OARDB6 can mark the trait of wool diameter, and revealed near these may exist the QTL which control the trait of wool diameter.(3)OARDB6 linkage closely with the wool intensity and can mark the trait of wool intensity.This study lay a foundation for location of sheep main economic trait QTL, and offer candidate-gene for molecular genetics, breading and marker associate selection at the molecular-level in Chinese Merino(Xinjiang army westland reclamation model)... |
