Sperm capacitation is an essential mature process before mammalian fertilization. Makingsperm capacitated in vitro is an important prerequisite to complete fertilization in vitro. Inthis paper, the Effects of Hypotaurine and Hyaluronidase on Wapiti(Cervus elaphus) SpermCapacitation In Vitro were studied, in order to choose a set of perfect and feasible protocol ofhypotaurine and hyaluronidase.To make the wapiti sperm capacitated in vitro, we selected and used the method of sperm-up. By making the rate of acrosome reaction as a main index, and the sperm energy, theviability as auxiliary indexes, it were carried out to compare the effects of wapiti spermcapacitation in vitro under different culture medium with hypotaurine and hyaluronidase. Theseresults showed that:.1,The effects of wapiti sperm capacitation in vitro were increased with 30~200μmol/Lhypotaurine in BO, the most suitable concentration for hypotaurine was 100μmol/L.The effectsof wapiti sperm capacitation in vitro were increased with 30~200μmol/L hypotaurine inTALP,the most suitable concentration for hypotaurine was 100μmol/L.Using TALP withhypotaurine was obviously higher than BO(p<0.05).2,The effects of wapiti sperm capacitation in vitro were increased with 0.5~1.5g/Lhyaluronidase in BO, the most suitable concentration for hyaluronidase was 70μmol/L. Theeffects of wapiti sperm capacitation in vitro were increased with 1g/L hyaluronidase inTALP,the most suitable concentration for hyaluronidase was 1g/L. Using BO withhyaluronidase was obviously higher than TALP(p<0.05).3,Viability were increase with 30~200μmol/L hypotaurine in BO. Viability were increasewith 30~200μmol/L hypotaurine in TALP.4,Hyaluronidase had no effects on viability of wapiti sperm.
