Red deer(Cervus elaphus) is listed as a second category wildlife protected by China. In the Heilongjiang Province belong to northeast China subspecies (Cervus elaphus xanthopygus). The red deer is of higher economic value and conservation signification. It's status of Chinese bio-diversity is very important. Habitat fragmentation on red deer in Wandashan Mountains is more and more grievous. At present, the study of heredity on northeast China red deer is not developed. The lack of heredity data will restrict the conservation of red deer to a large extent.In the study, we analyzed genetic diversity of red deer in the Wandashan Mountains. We use microsatillites DNA markers. Twenty microsatellites from California elk (Cervus elaphus canadensis) and six microsatellites from black-tailed deer (O. hemionus). Eight polymorphic microsatellites loci were found. The 8 microsatellites loci were used to amplifiy 150 fecal samples. 50 of them were amplified successfully. We identified individuals and tested the genetic diversity in 50 red deer fecal samples using these eight polymorphic microsatellites loci. The PCR amplified products of microsatellites loci were detected by denatured polyacry lamide gel electrophoresis. The results showed these fecal samples belonged to 31 different individual; we calculated the gene heterozygosity, individual identification potential, polymorphism information content, and effective number of alleles of each locus. The observe heterozygosity(Ho) of 8 loci ranged from 0.3871 to 0.8065, averaging 0.5928 per locus; the expectation heterozygosity(He) of 8 loci ranged from 0.3314 to 0.8174, averaging 0.6244 per locus; the effective number of alleles(Ne) of 8 loci ranged from 1.4957 to 5.4765, 3.1491 alleles per locus on average; The polymorphism information content(PIC) of 8 loci ranged from 0.7927 to 0.2765, averaging 0.5388 per locus. The total discrimination power (TDP) of 8 loci was 99.99%. The results of Sry-PCR amplified analysis of 31 samples, showed that there 19 male and 12 female. In the study area, the sexual ratio of natural population is 0.63:1(19:12).
