Seven sweet corn(Wanyu 10,Wanyu 14,Wanyu 15,Wantian No.1,Lilifeng,Shuangfu, Jinfei)were treat by three priming treatments(Sand priming,H2O2 priming,Sand+SNP compound priming)which including four disposals,and their seed vigor were tested by ways of seedling growth test,accelerated ageing test,cold test,electrical conductivity test, and simulated field seedling growth test,etc.More suitable and effective methods for improve seed vigor of seven sweet corn were found by analyzed the effect of different disposals to the sweet corn seeds.On the base of it,the change of three enzymes(TTC, POD,Apase)activation with the feasible disposals were researched to discuss the effect of priming methods to enzymes activation.And put up isozyme electrophoresis analyse to make more research to discuss the effect of priming methods to physiological and biochemical mechanism.1.Notably or significantly positive correlation were found between the field emergence percentage and the vigor target including germination energy,germination percentage and germination index in seedling growth test,simulated field seedling growth test and cold test.According to the results of this experiment,the seedling growth test,cold test and simulated field seedling growth test were the most proper methods for testing the seed vigor of sweet corn.2.Generally speaking,5.7%and 7.4%were the better disposals which could improve the sweet corn's vigor.Mostly results of accelerated ageing test,cold test,simulated field seedling growth test were higer than comparison,but the significantly was not notably. Three enzymes(TTC,POD,Apase)activation were higer than comparison.The Apase color of sand priming was darker than comparison,the number of POD band was more than comparison.3.0.75%was the best concentration in all H2O2 priming,which most of vigor index were higer than dry seed and water soaking.In accelerated ageing test,the results of wanyu14 and wanyu15 were high significantly,effect of 0.50%and 0.75%was notably or high significantly higer than comparison in three enzymes(TTC,POD,Apase)activation. the number of POD band was more than comparison. 4.Along with the increase of SNP concentration,the vigor index of seedling growth test,accelerated ageing test,cold test was first raise and then descendant.Of all the Sand+SNP compound priming disposals,2.0mmol/L was the best concentration,which most of vigor index was higer than dry seed and sand priming.Effect to enzymes activation of 1.5mmol/L and 2.0mmol/L were notably,part of them was lower then sand priming.The Apase color of Sand+SNP priming was darker than dry seed comparison,but more undertone than sand priming.the number of POD band was more than comparison and more undertone than sand priming. |
