| Sugarcane (Saccharum officinarum L.) is the most important sugar crops and an important energy crop in China. Sugarcane stem borer and Ceratovacuna lanigera Zehnther are the main pests of sugarcane, which not only make sugarcane yields loss but also make sucrose content decline. So, breeding insect-resistant sugarcane varieties by using plant genetic engineering techniques is the most effective ways.The galanthus nivalis agglutinin (GNA) is one kind of plant mannose combine agglutinin, it can integrate on the glycoprotein acceptor of insect enteron epithelial cell, and produces partial or system poisons to the insect and suppresses its growth. Therefore, transferring the GNA gene into sugarcane and making it express effectively can enhance the ability of vermin- resistance in sugarcane.A plant expression vector EprG, which the GNA gene was driven by the promoter rbcS, was constructed. Then the EprG was introduced into the super-poison Agrobacterium tumerfaciens EHA105 through the"Freezing and thawing"method and used to transform sugarcane embryonic calli by Agrobacterium tumerfaciens-mediated method. The transformed shoots were screened under the selective pressure of PPT and 53 resistant shoots were obtained. Two transgenic plants were confirmed by using the method of PCR from 10 resistant shoots. It primary show that the GNA gene was integrated into the sugarcane genome. |
